多肽添加水平对刺参生长性能、免疫及肠道功能基因表达的影响 被引量:1

Effects of Peptide Supplemental Levels on Growth Performance,Immunity and Expression of Intestine Functional Genes in Sea Cucumber(Apostichopus japonicus)

摘要 本试验旨在探究多肽添加水平对刺参生长性能、免疫及肠道功能基因表达的影响。试验选择300头刺参[体重(34.40±0.23)g)],根据不同多肽添加水平(0、5%、10%、15%、20%),分为5组(D1、D2、D3、D4、D5组),每组3个重复,每个重复20头刺参,探讨多肽对生长性能及肠道功能基因(腱生蛋白、胶原蛋白α2、整合素αV、整合素βL、p105、p50、rel和lys)表达的影响。试验期60 d。结果表明:D4组的刺参特定生长率(SGR)显著高于其他组(P<0.05),较D1组提高了136%,摄食率(IR)低于其他各组(P>0.05),较D1组降低了25%。体腔液中总抗氧化能力(T-AOC)、超氧化物歧化酶(SOD)、酸性磷酸酶(ACP)和碱性磷酸酶(AKP)活性随着多肽添加水平的增加呈现升高趋势。D2、D3、D4组腱生胶原蛋白、胶原蛋白α2、整合素αV、整合素βL基因在刺参肠道内相对表达量均显著高于D1组(P<0.05),在D4组达到表达高峰。p105、p50、lys基因相对表达量随着多肽添加水平的增加呈现持续升高趋势。由此可见,饲粮中添加10%~15%多肽能显著提高刺参的SGR,降低饵料FR,同时上调刺参肠道内生长类基因(腱生蛋白、胶原蛋白α2、整合素αV和整合素βL)的表达以及免疫类基因(p105、p50和lys)的表达,从而促进刺参生长,提高刺参自身免疫力。 This experiment was conducted to determine the effects of peptide supplemental levels on growth performance,immunity and expression of intestine functional genes in sea cucumber(Apostichopus japonicas).A total of 300 sea cucumbers with an initial body weight of(34.40±0.23)g were selected,according to peptide supplemental levels(0,5%,10%,15%and 20%),divided into 5 groups(D1,D2,D3,D4 and D5 groups),and each group had 3 replicates,each replicate had 20 sea cucumbers.The test was investigated the effects of polypeptides on growth performance and expression of intestinal functional genes(tendinogenic protein,collagenα2,integrinαV,integrinβL,p105,p50,rel and lys).The experiment lasted for 60 d.The results showed as follows:specific growth rate(SGR)of sea cucumbers in D4 group was significantly enhanced(P<0.05)and increased by 136%than that in D1 group;feeding rate(FR)was lower than that in other groups(P>0.05),and reduced by 25%.The antioxidant capacity(T-AOC),activities of superoxide capacity(SOD),acid phosphatase(ACP)and alkaline phosphatase(AKP)in coelomic fluid were increased with the increasing peptide supplement level.The expression levels of tenascin,collagenα2,integrinαV,intergrinβL genes in sea cucumbers in D2,D3 and D4 groups were significantly higher than those in the D1 group(P<0.05),and reached the peak in D4 group.The expression levels of p105,p50,rel and lys genes were increased with increasing amounts of peptide supplements.It is concluded that dietary 10%to 15%peptide can significantly increase the SGR of sea cucumber and decrease the FR.The expression of intestinal growth genes(tenascin gene,collagenα2 gene,integrinαV and intergrinβL)and immune genes(p105,p50,and lys)are up-regulated so as to promote the growth of sea cucumber and improve its immunity.
