摘要
目的研究夜仁合有效部位改善睡眠的量效关系、并通过成分分析和脑内神经递质变化,探讨夜仁合有效部位改善睡眠的物质基础和作用机制。方法以雄性ICR小鼠为实验动物,首先采用戊巴比妥钠睡眠协同实验进行夜仁合有效部位改善睡眠作用的量效关系研究;其次,采用高效液相色谱法(HPLC),确定夜仁合有效部位中具有改善睡眠作用的成分及含量;最后,以最佳量效开展夜仁合有效部位改善睡眠作用的机制研究。结果夜仁合有效部位0.15 g·kg^(-1)组可显著缩短睡眠潜伏期(P<0.05),0.3 g·kg^(-1)组可以极显著缩短睡眠潜伏期(P<0.01)和显著延长睡眠时间(P<0.01)。夜仁合有效部位中具有改善睡眠作用的有效成分为五味子甲素含量1.08%、五味子乙素含量0.92%、斯皮诺素含量2.36%、槲皮苷含量0.07%,4种成分总含量为4.43%。0.3 g·kg^(-1)组能明显提高小鼠下丘脑内5-羟色胺(5-HT)和5-羟吲哚乙酸(5-H1AA)的含量,降低去甲肾上腺素(NE)的含量。结论夜仁合有效部位(五味子甲素含量1.08%、五味子乙素含量0.92%、斯皮诺素含量2.36%、槲皮苷含量0.07%)在一定剂量内具有改善睡眠的作用,最佳量效为0.3 g·kg^(-1),改善睡眠的作用机制与中枢神经递质(5-HT、5-HIAA和NE)调节有关。
Objective To study the dose-effect relationship of the active parts of YERENHE in improving sleep,and to explore the material basis and mechanism of action for the effective parts of YERENHE to improve sleep through component analysis and changes in brain neuro-transmitters.Methods Male ICR mice were used in the current experiments.First,the pentobarbital sodium sleep synergy experiment was used to study the dose-effect relationship of the active parts of YERENHE in improving sleep.Then,high performance liquid chromatography(HPLC)was used to determine YERENHE.The ingredients and content of the effective parts of the combination have the effect of improving sleep.Finally,the mechanism of the effective parts of YERENHE combination to improve sleep is carried out with the best dose effect.Results The effective part of YERENHE 0.15 g·kg^(-1)group can significantly shorten the sleep latency(P<0.05),and the 0.3 g·kg^(-1)group can significantly shorten the sleep latency(P<0.01)and prolong sleep time(P<0.01).The active ingredients in the effective parts of YERENHE that can improve sleep are jujuboside A 1.08%,jujuboside B 0.92%,spinosin 2.36%,quercetrin 0.07%.The total content of the 4 ingredients is 4.43%.The 0.3 g·kg^(-1)group significantly increases the content of 5-HT and 5-H1 AA in the hypothalamus of mice,and reduces the content of NE.Conclusion The effective part of YERENHE(jujuboside A 1.08%,jujuboside B 0.92%,spinosin 2.36%and quercetrin 0.07%)can improve sleep in a certain dose.The mechanism of sleep improvement is related to central neurotransmitters(5-HT、5-HIAA and NE)regulation.
