摘要
建立高效液相色谱法同时测定清胃润肠合剂中芒果苷、盐酸巴马汀、盐酸小檗碱、甘草酸含量的方法。采用Welchrom■C_(18)(4.6 mm×250 mm,5μm)色谱柱,流动相为乙腈-0.05 mol/L磷酸二氢钾溶液(用磷酸调节pH值至3.5),梯度洗脱,流速1.0 mL/min,柱温30℃,进样量10μL,检测波长258 nm。结果表明,芒果苷、盐酸巴马汀、盐酸小檗碱、甘草酸分别在13.600~217.550μg/mL、9.410~150.560μg/mL、35.670~570.690μg/mL、40.910~654.560μg/mL范围内线性关系均良好(r>0.999 0),平均加样回收率(n=6)分别为102.88%、102.60%、99.50%、100.77%,RSD分别为0.49%、0.91%、0.49%、0.41%。该方法快速简便、精密度好、灵敏度高,可用于清胃润肠合剂的质量控制。
To establish a method for the simultaneous content determination of mangiferin,palmatine chloride,berberine hydrochloride and glycyrrhizic acid in Qingwei Runchang mixture by HPLC,the determination was carried out with Welchrom■C_(18) column(4.6 mm×250 mm,5 μm). The mobile phase was consisted of acetonitrile(A) and0.05 mol·L;potassium dihydrogen phosphate solution(B,adjusted pH value to 3.5 by phosphoric acid)with gradient elution at a flow rate of 1.0 mL·min-1,sample volume was 10 μL,and the detection wavelength was 258 nm. The results showed that mangiferin,palmatine chloride,berberine hydrochloride and glycyrrhizic acid had good linear relationship within the range of 13.600-217.550 μg·mL^(-1),9.410-150.560 μg·mL^(-1),35.670-570.690 μg·mL^(-1),40.910-654.560 μg·mL^(-1),respectively(r>0.999 0). The average recovery(n=6)was 102.88%,102.60%,99.50%and 100.77% with RSD of 0.49%,0.91%,0.49% and 0.41%. The method can be served as a scientific and effective way for quality control of Qingwei Runchang mixture.
