摘要
目的为了提高食管鳞癌患者的生存期,探讨CNTNAP3B基因单核苷酸多态性(SNP)与食管鳞癌患者生存期(OS)的关联。方法选择高发地区医院的199位食管鳞癌患者的癌组织为研究对象,使用bcftools软件识别SNP,并利用ANNOVAR软件对多态性位点进行功能注释。采用液相色谱—质谱技术对199例食管鳞癌的癌组织样本进行分析,并采用SPSS 21.0分析SNP rs3739620位点基因型与食管鳞癌患者OS的关系。结果单个SNP位点分析时,发现C/T基因型比C/C基因型生存率高,T/T基因型比C/T基因型生存率高。统计学分析显示SNP不同基因型的食管鳞癌患者的生存有显著差异(P<0.02)。CNTNAP3B基因rs3739620位点基因型影响MRPL18蛋白表达,以T/T基因型蛋白表达量最低,C/C基因型蛋白表达量最高,差异有统计学意义(P<0.001,bate为-0.32)。结论CNTNAP3B基因rs3739620位点T/T基因型生存期最好,可能与通过CNTNAP3B基因影响MRPL18蛋白表达有关,对食管鳞癌患者预后生存具有一定的临床指导意义。
Objective To improve the survival of patients with esophageal squamous cell carcinoma,the association between single nucleotide polymorphism(SNP)of CNTNAP3B gene and survival(OS)of patients with esophageal squamous cell carcinoma was investigated.Methods The cancer tissues of 199 patients with esophageal squamous cell carcinoma from hospitals in high incidence areas were selected.SNP was identified by bcftools software,and functional annotation of polymorphic loci was performed by ANNOVAR software.The tissue samples of the 199 cases were analyzed by liquid chromatography-mass spectrometry.SPSS 21.0 was used to analyze the relationship between SNP genotype and OS in patients with esophageal squamous cell carcinoma.Results Single SNP analysis showed that C/T genotype had a higher survival rate than C/C genotype,and T/T genotype had a higher survival rate than C/T genotype.Statistical analysis showed that there was a significant difference in survival between different SNP genotypes and esophageal squamous cell carcinoma patients(P<0.02).CNTNAP3B locus rs3739620 different genotypes affected MRPL18 protein expression,especially T/T genotype protein expression was the lowest,C/C genotype protein expression was the highest,the difference was statistically significant(P<0.001,BATE was-0.32).Conclusion CNTNAP3B gene rs3739620 locus T/T genotype has the best survival,and CNTNAP3B gene affects the down-regulation of MRPL18 protein expression,which has certain clinical guiding significance for the prognosis and survival of patients with esophageal squamous cell carcinoma.
