摘要
目的建立低共熔溶剂(deep eutectic solvent,DES)-超高效液相色谱-串联质谱法(ultra performance liquid chromatography-tandem mass spectrometry,UPLC-MS/MS)快速检测肉制品、水产品中3种非法添加工业染料(刚果红、酸性金黄、红2G)的分析方法。方法样品经水稀释,微量DES分散萃取,乙腈复溶。三重四极杆电喷雾离子源电离-多反应监测(multiple reaction monitoring,MRM)-负离子监测模式检测,外标法定量。结果刚果红和红2G在0.2~20.0μg/kg范围内、酸性金黄在0.02~2.00μg/kg范围内线性关系良好,相关系数均大于0.99。方法的检出限分别为0.12、0.26、0.01μg/kg,回收率为93.25%~101.36%,相对标准偏差为1.27%~4.06%。结论低共熔溶剂对刚果红、酸性金黄、红2G的萃取效果佳,可以解决传统操作步骤烦琐的问题。本方法快速、简便、精准,适用于肉制品、水产品中3种非法添加工业染料刚果红、酸性金黄、红2G的检测。
Objective To establish a rapid method for determination of 3 kinds of illegally added industrial dyes(Congo red,methaniol yellow,red 2G)in meat and aquatic product by ultra performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS)with deep euectic solvent(DES).Methods The samples were dissolved by water,dispersion extracted by microscale deep euectic solvent,and dissoluted by acetonitrile.Triple quadrupole electrospray ion source ionization-multiple reaction monitoring(MRM)-anion monitoring mode was used for detection,external standard method was used for quantitative analysis.Results The linear relationships of Congo red,red 2G in the ranges of 0.2-20.0μg/kg and metanil yellow in the ranges of 0.02-2.00μg/kg were good,respectively,and the correlation coefficients were greater than 0.99.The limits of detection of the method were 0.12,0.26 and 0.01μg/kg,respectively.The recovery rates were 93.25%-101.36%,and relative standard deviations were 1.27%-4.06%.Conclusion DES has a good extraction effect of Congo red,red 2G and metanil yellow,which can solve the problem of complicated tradittional operation steps.This method is rapid,simple and accurate,and is suitable for determination of 3 kinds of illegally added industrial dyes in meat and aquatic products.
