摘要
目的探讨成纤维生长因子17(FGF17)在食管癌组织中的表达,并分析其与食管癌患者临床病理特征、预后的关系。方法收集2016年1月至2021年12月在郑州大学第一附属医院行外科手术切除的174例食管癌患者的肿瘤组织及对应的癌旁正常食管组织。用实时定量反转录聚合酶链反应(RT-qPCR)技术检测并比较不同食管组织中FGF17基因的表达,分析其与食管癌患者临床病理特征的关系预后的关系。用免疫组化染色和组织芯片技术检测食管癌中106例食管癌和61例相邻癌旁正常食管组织中FGF17蛋白的表达,分析其与患者预后的关系。结果RT-qPCR结果显示食管癌组织中FGF17基因表达水平(8.36±0.51)高于癌旁正常食管组织(8.07±0.44),差异有统计学意义(t=10.850,P<0.001);食管癌组织中FGF17基因表达水平与食管癌患者T分期、N分期有关(χ^(2)=7.436,P=0.006;χ^(2)=8.064,P=0.005)。免疫组化染色结果显示食管癌组织中FGF17蛋白阳性表达率为74.53%(79/106),高于癌旁正常食管组织的37.70%(23/61),差异有统计学意义(χ^(2)=22.080,P<0.001);FGF17蛋白高表达患者总体存活率明显低于FGF17蛋白低表达患者,差异有统计学意义(χ^(2)=7.577,P=0.006)。结论FGF17在食管癌组织中存在高表达,表达水平随食管癌疾病进展升高,且高表达预示着较差的预后。
Objective To explore the expression of fibroblast growth factor 17(FGF17)in the esophageal cancer tissues,and to analyze the correlation between the expression level and clinical pathological characteristics and prognosis of patients with esophageal cancer.Methods Cancer tissues and corresponding para-cancer tissues were collected from 174 patients with esophageal cancer who underwent surgical resection in the First Affiliated Hospital of Zhengzhou University from January 2016 to December 2021.Real-time quantitative reverse transcription polymerase chain reaction(RT-qPCR)was used to test FGF17 gene expression in the cancer tissues and corresponding para-cancer tissues of 174 esophageal cancer patients.The correlation between the expression level clinical and the pathological characteristics was analyzed.The immunohistochemistry assay and the tissue micro-array were utilized to examine the FGF17 expression in the esophageal cancer tissues as well as adjacent normal tissues.The correlation between the expression level clinical and the prognosis was analyzed.Results The result of RT-qPCR showed that the expression of FGF17 in the esophageal cancer tissues(8.36±0.51)was higher than that in adjacent normal tissues(8.07±0.44),the difference between the two groups was statistically significant(t=10.850,P<0.001).The expression levels of FGF17 were observably associated with T and N stages(χ^(2)=7.436,P=0.006;χ^(2)=8.064,P=0.005).Immunohistochemical results showed that the positive expression rate of FGF17 in the esophageal cancer tissues was 74.53%(79/106),which was higher than that in para-cancer tissues 37.70%(23/61),the difference was statistically significant(χ^(2)=22.080,P<0.001).The overall survival rate of patients with high FGF17 expression was remarkably lower than that of patients with low FGF17 expression(χ^(2)=7.577,P=0.006).Conclusion The expression of FGF17 is higher in esophageal cancer tissues,and the expression level increases with the progression of esophageal cancer,and the high expression indicates a poor prognosis.
