摘要
目的针灸联合加减二仙汤对卵巢储备功能减退(DOR)大鼠血清中抗苗勒激素(AMH)的影响。探究针灸联合加减二仙汤方对DOR的预防、调节和改善作用,并为临床上针药并治DOR提供理论基础。方法从60只清洁级雌性SD大鼠中随机选取10只设置成空白组,剩余的50只大鼠以环磷酰胺(CTX)腹腔注射制作出卵巢储备功能下降的大鼠模型,再将其随机分成模型组、中药组(加减二仙汤)、针刺组、中药联合针刺组、西药组(DHEA)。采用放射免疫法检测每组大鼠血清中AMH的水平,HE染色镜下观察各组大鼠卵巢组织形态。结果与空白组比较,模型组大鼠卵泡数目及颗粒细胞层数减少,闭锁卵泡数目增多,AMH显著降低(P<0.01);与模型组比较,中药联合针刺组原始卵泡数目增加,闭锁卵泡数目减少,颗粒细胞层数增多,AMH显著升高(P<0.01)。结论针灸联合二仙汤对卵巢损伤具有一定的保护预防作用,可促进卵泡发育,减缓闭锁卵泡的生长,并且有效增加DOR大鼠血清中的AMH,进一步提高卵巢功能。
Objective The effect of acupuncture combined with disar soup on anti-Miaole hormone(AMH)in ovarian reserve function reduction(DOR)rat serum.To explore the prevention,regulation and improvement of DOR by combining acupuncture with the addition and subtraction of two fairy soups.It also provides a theoretical basis for clinical injection and treatment of DOR.Methods From 60 clean female SD rats,10 were randomly selected into blank groups,and the remaining 50 rats produced a mouse model with reduced ovarian reserve function by injection of cyclophosphamide(CTX)abdominal cavity,and then randomly divided into model groups,Chinese medicine groups(plus minus two fairy soup),needles,Chinese medicine joint needles,Western medicine groups(DHEA).The level of AMH in each group of rats was detected by radioimmune method,and the ovarian tissue pattern of each group was observed under the HE staining mirror.Results Compared with blank group,the number of follicles and granulosa cell layers in model group the number of atretic follicles increased,and AMH was significantly decreased(P<0.01).Compared with model group,the number of original follicles,the number of atretic follicles,the number of granulosa cell layers and AMH were increased significantly in TCM combined with acupuncture group(P<0.01).Conclusion Acupuncture combined with Erxian Decoction has a certain protective and preventive effect on ovarian injury,which can promote the development of follicles,slow down the growth of atretic follicles,and effectively increase AMH in serum of DOR rats to further improve ovarian function.
