网络药理学研究玉泉丸治疗2型糖尿病的机制 被引量:1

Network Pharmacological Study on the Mechanism of the Yuquan Pill in Treating Type 2 Diabetes Mellitus

摘要 目的:基于网络药理学研究玉泉丸治疗2型糖尿病的机制。方法:在TCMSP和ETCM数据库中筛选玉泉丸的有效成分和靶点,使用GeneCards数据库汇总2型糖尿病(Type 2 diabetes mellitus)的靶点,与玉泉丸的靶点取交集得到共有靶点。导入String构建靶点互作网络,借助Cytoscape 3.9.0构建“成分—靶点”图,以“Degree值大于2倍平均值”以及“Betweenness Centrality值大于2倍平均值”为条件筛选关键靶点,导入Metascape进行GO和KEGG富集。结果:玉泉丸的有效成分为166个,其中葛根4个,天花粉2个,五味子8个,甘草92个,地黄12个,麦冬48个;2型糖尿病的靶点有1449个;玉泉丸和2型糖尿病的共有靶点有304个,其中关键靶点有25个;GO富集得到BP 687条,MF 31条,CC 21条通路,共计739条信号通路;KEGG得到139条通路。结论:玉泉丸中麦冬地黄的共有成分腺苷和西托糖苷、葛根甘草的共有成分芒柄花黄素、葛根中的大豆苷元、天花粉中的固醇、麦冬中的麦冬酮B、地黄中的水苏糖、五味子中的五味子素C、甘草中的异鼠李素和异黄酮醇,通过作用于AKT1、TNF、VEGFA、EGFR、STAT3、MAPK1、APP和GSK3B等关键靶点,激活HIF-1信号通路、胰岛素抵抗和cAMP信号通路,发挥治疗2型糖尿病的作用。 Objective:The mechanism of the Yuquan pill in treating type 2 diabetes is studied by network pharmacology.Methods:With the help of TCMSP and ETCM database,the active ingredients and targets of the Yuquan pill were retrieved.Firstly,“Type 2 diabetes mellitus”targets are summarized by using GeneCards database.Then,the targets intersect with the target of the Yuquan Pill to obtain the common target.With the help of importing String to build the target interaction network,and building the“component-target”diagram using Cytoscape 3.9.0.,the key targets were screened under the conditions that“dgree value is greater than 2 times the average value”and“betweenness centrality value is greater than 2 times the average value”,and then Metascape was imported for GO and KEGG enrichment.Results:There are 166 active ingredients in the Yuquan Pill,including 4 pueraria lobata,2 trichosanthin,8 schisandra chinensis,92 licorice,12 rehmannia glutinosa and 48 ophiopogon japonicus;1449 targets for type 2 diabetes.304 targets were found in the Yuquan pill and type 2 diabetes,including 25 target targets.739 signal pathways were enriched by GO,including 687 BP,31 MF,21 CC;139 signal pathways were enriched by KEGG.Conclusion:Adenosine and sitogluside in ophiopogon japonicus and rehmannia glutinosa,formononetin in pueraria lobata and licorice,daidzein in pueraria lobata,schottenol in trichosanthin,ophiopogonanone B in ophiopogon japonicus,stachyose in rehmannia glutinosa,wuweizisu C in schisandra chinensis,isorhamnetin and isolicoflavonol in licorice can activate HIF-1 signaling pathway,insulin resistance and cAMP signaling pathway by acting on key targets such as AKT1,TNF,VEGFA,EGFR,STAT3,MAPK1,APP and GSK3B,and play a role in the treatment of type 2 diabetes.
