摘要
目的从网络药理学角度研究黄精的主要活性成分及其治疗2型糖尿病的可能作用机制。方法从TCMSP数据库中提取黄精的化学成分,以口服生物利用度(OB)≥30%和类药性(DL)≥0.18筛选活性成分,通过该数据库提取活性成分作用靶点,以DisGeNet及OMIM在线分析平台挖掘疾病相关靶点,最后采用Perl语言与R语言分析靶点关联的主要生物通路,获取黄精活性成分的多维角度抗2型糖尿病的作用机制。结果通过筛选获得DFV、黄芩素、3’-甲氧基黄豆苷元、β-谷甾醇、甲基原薯蓣素、甘草素、薯蓣皂甙元、谷甾醇、4’,5-二羟基黄酮等成分等12个主要活性成分;这些成分可作用于靶点68个,其中核心靶点有AKT1、JUN、FOS、VEGFA、CASP3、MMP9等;这些成分可作用炎症感染(乙型肝炎、人类巨细胞病毒感染)、癌症通路、AGE-RAGE信号通路、TNF信号通路、脂质和动脉粥样硬化、非酒精性脂肪肝、胰岛素抵抗、雌激素信号通路、IL-17信号通路、HIF-1信号通路等。进一步发现抗2型糖尿病作用黄精中主要活性成分可通过“多成分-多靶点-多通路”来发挥治疗糖尿病的作用。结论从网络药理学预测了黄精抗2型糖尿病的主要活性成分和多维抗2型糖尿病药理作用机制,为进一步探讨其作用机制奠定了基础。
Objective: To study the main active ingredients of Polygonatum chinensis and its possible mechanism of action in the treatment of diabetes patients(Ⅱ) from the perspective of network pharmacology. Methods: The chemical constituents of Polygonatum chinensis were extracted from the TCMSP database, and the active ingredients were screened with oral bioavailability(OB) ≥ 30% and drug-like properties(DL) ≥ 0.18. The target of active ingredients was extracted from this database, and the disease-related targets were mined by DisGeNet and OMIM online analysis platform. Finally, Perl language and R language were used to analyze the main biological pathways associated with the targets, and to obtain the multi-dimensional anti-type Ⅱ diabetes mechanism of the active ingredients of Polygonatum chinensis. Results: 12 main active ingredients such DFV, baicalein, 3’-methoxydaidzein, β-sitosterol, methylprotodioscin, liquiritigenin, diosgenin, sitosterol, 4’, 5-dihydroxyflavone, etc. were obtained by screening. These ingredients could act on 68 targets, of which the core targets were AKT1, JUN, FOS, VEGFA, CASP3, MMP9, etc.. These ingredients could act on inflammatory infections(hepatitis B and human giant cells virus infection), cancer pathway, AGE-RAGE signaling pathway, TNF signaling pathway, lipids and atherosclerosis, non-alcoholic fatty liver disease, insulin resistance, estrogen signaling pathway, IL-17 signaling pathway, HIF-1 signaling pathway. It was further found the main active ingredients of Polygonatum chinensis could play a role in the treatment of diabetes through "multi-ingredients-multi-target-multi-pathway". Conclusion: The main active ingredients of Polygonatum in anti-type Ⅱ diabetes patients and the pharmacological mechanism of multi-dimensional anti-type Ⅱ diabetes patients are predicted from the network pharmacology, which lays a foundation for further exploration of its mechanism of action.
