摘要
目的通过对癌症和肿瘤基因图谱(TCGA)和GTEx数据库中胶质瘤和正常脑组织的基因测序数据进行挖掘,探讨GAS2L3在正常脑组织和不同等级的胶质瘤中差异及其可能对胶质瘤细胞的影响,以期建立胶质瘤治疗的新靶点。方法通过基因表达谱交互分析(GEPIA)网站分析TCGA和GTEx数据库中GAS2L3在高级别胶质瘤和低级别胶质瘤与正常脑组织中的表达差异。从TCGA数据库中获取702例胶质瘤组织的RNA测序数据与临床信息,首先分析GAS2L3与胶质瘤的等级与预后的相关性;其次通过单因素与多因素的回归分析分析GAS2L3的风险比;最后通过相关性分析计算GAS2L3与增殖、侵袭、凋亡和血管形成的相关性。在以上数据中,按照log FC>1且P<0.05的标准筛选GAS2L3高表达与GAS2L3低表达胶质瘤中的差异表达基因,并通过基因本体(GO)和京都基因与基因组百科全书(KEGG)分析其在胶质瘤中的调控方式,基因集富集分析(GSEA)其对细胞信号通路的影响。结果通过GEPIA网站分析发现GAS2L3表达量在低级别胶质瘤与正常脑组织无明显差异,但高表达于胶质母细胞瘤中。GAS2L3在TCGA胶质瘤数据中与胶质瘤的等级正相关,与胶质瘤的预后负相关。单因素与多因素的回归分析发现GAS2L3是胶质瘤预后的不良因素。相关性分析显示GAS2L3与胶质瘤的增殖明显相关。在GAS2L3高表达与低表达胶质瘤中发现2887个差异基因,GO和KEGG富集其影响的信号通路,GSEA分析发现GAS2L3激活细胞周期信号通路。结论GAS2L3基因可能在胶质瘤的诊断和临床治疗中有重要的意义。
Objective To explore the expression difference of growth arrest-specific 2-like protein 3(GAS2L3)in normal brain tissue and different grades of glioma and its possible effect on glioma cells.In this study,the gene sequencing data of glioma and normal brain tissue were excavated from The Cancer Genome Atlas(TCGA)and Genotype-Tissue Expression(GTEx)to find new targets for the treatment of glioma.Methods The difference of GAS2L3 expression in highgrade gliomas,low-grade gliomas and paratumoral tissues in TCGA and GTEx databases was analyzed by Gene Expression Profiling Interactive Analysis(GEPIA)website.The RNA sequences and clinical information of 702 glioma tissues were obtained from TCGA database.First,the correlation between GAS2L3 and glioma grade and prognosis was analyzed.Second,the risk ratio of GAS2L3 was analyzed by univariate and multivariate regression analysis.At last,the correlation between GAS2L3 and proliferation,invasion,apoptosis and angiogenesis was calculated.In glioma database,the absolute value of logFC>1 and P<0.05 were used as the criteria to screen the differentially expressed genes in gliomas with high expression of GAS2L3 and low expression of GAS2L3.The regulation patterns of the related genes in glioma were analyzed by Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG).Gene set enrichment analysis(GSEA)was used to analyze its effect on cell signaling pathway.Results Through the analysis of GEPIA website,it was found that there was no significant difference in the expression of GAS2L3 between low-grade gliomas and normal brain tissues,but it was highly expressed in glioblastoma.GAS2L3 was positively correlated with the grade of glioma and negatively correlated with the prognosis of glioma in TCGA glioma data.Univariate and multivariate regression analysis showed that GAS2L3 was a poor prognostic factor for glioma.Correlation analysis showed that GAS2L3 was significantly correlated with glioma proliferation.2887 differential genes were found in gliomas with high and low expression of GAS2L3.GO and KEGG enriched the signal pathways affected by them.GSEA analysis found that GAS2L3 activated the cell cycle signal pathway.Conclusion GAS2L gene may play an important role in the diagnosis and clinical treatment of gliomas.
