基于lncRNA MALAT1/TGF-β通路调控EMT探讨白花蛇舌草抑制大肠癌细胞的迁移和侵袭 被引量:4

摘要 目的探究白花蛇舌草(HDW)通过长非编码RNA肺癌转移相关转录本1(lncRNA MALAT1)/转化生长因子β(TGF-β)通路调控上皮间质转化(EMT)对大肠癌细胞的迁移和侵袭的抑制作用。方法取对数生长期的SW620细胞分为0 mg/mL组、0.5 mg/mL组、1 mg/mL组、2 mg/mL组,各组分别加入0、0.5、1、2 mg/mL HDW药液干预24 h,24 h后采用倒置显微镜观察细胞数量、形态变化,CCK-8法检测细胞活力,划痕实验和Transwell检测细胞迁移和侵袭能力,RT-qPCR检测lncRNA MALAT1相对表达量,Western blot检测TGF-β、Smad4、Vimentin、E-cadherin、N-cadherin蛋白表达。结果与0 mg/mL组比较,各药物组随着HDW药物浓度的增加,SW620细胞数量减少,形态皱缩变圆,细胞活力逐渐下降(P<0.05),细胞迁移和侵袭能力逐渐降低,细胞lncRNA MALAT1相对表达量逐渐下调(P<0.05),细胞TGF-β、Smad4、Vimentin、N-cadherin蛋白表达逐渐降低(P<0.05),E-cadherin蛋白表达逐渐增高(P<0.05)。结论HDW可通过抑制SW620细胞lncRNA MALAT1/TGF-β通路调控EMT,进而抑制大肠癌SW620细胞的迁移和侵袭。 Objective:To explore the inhibitory effect of Hedyotis diffusa Willd.(HDW)on the migration and invasion of colorectal cancer cells by regulating epithelial-mesenchymal transition(EMT)through long non-coding RNA metastasis-associated lung adenocarcinoma transcript 1(lncRNA MALAT1)/transforming growth factor-β(TGF-β).Methods:SW620 cells in logarithmic growth phase were divided into 0 mg/mL group,0.5 mg/mL group,1 mg/mL group and 2 mg/mL group.After 24 h intervention with 0 mg/mL,0.5 mg/mL,1 mg/mL and 2 mg/mL HDW solution,the number and morphology of cells were observed by inverted microscope;cell viability was detected by CCK-8 assay;the abilities of cell migration and invasion were separately analyzed by wound healing and Transwell assay;the relative expression of lncRNA MALAT1 was detected by RT-qPCR;the protein expressions of TGF-β,Smad4,Vimentin,E-cadherin and N-cadherin were detected by Western blot.Results:Compared with 0 mg/mL group,with the increase of HDW concentration,the number of SW620 cells in each drug group decreased,the cell morphology wrinkled and became round,and the cell viability decreased gradually(P<0.05);the ability of cell migration and invasion decreased gradually;the relative expression of lncRNA MALAT1 decreased gradually(P<0.05);the expressions of TGF-β,Smad4,Vimentin and N-cadherin protein decreased gradually(P<0.05),while the expression of E-cadherin protein increased gradually(P<0.05).Conclusion:HDW can regulate EMT by inhibiting lncRNA MALAT1/TGF-βpathway in SW620 cells,and then inhibit the migration and invasion of colorectal cancer SW620 cells.
