西洋参对抗生素相关性腹泻大鼠肠道菌群的影响研究 被引量:7

Effects of Xiyangshen(Panacis Quinquefolii Radix)on Intestinal Flora in Rats with Antibiotic-Associated Diarrhea

摘要 目的研究西洋参对抗生素相关性腹泻(Antibiotic-associated diarrhea,AAD)大鼠的干预作用,并分析其对肠道菌群的影响,初步探讨其作用机制。方法40只SD大鼠随机分为4组,每组10只。分别为空白对照组(Con),AAD模型组(Mod),西洋参低剂量组(XYS1),西洋参高剂量组(XYS2)。采用抗生素混合溶液灌胃方法建立抗生素相关性腹泻(AAD)大鼠模型,西洋参低、高剂量组分别给予西洋参水煎液1 g/(kg·d),2 g/(kg·d)灌胃治疗。评估大鼠腹泻状况,治疗结束后观察结肠组织变化,通过高通量测序法检测大鼠肠道菌群的变化,并进行功能预测分析。结果西洋参水煎液治疗后,腹泻症状及腹泻相关指标得到明显的改善结肠结构得到恢复。菌群分析结果显示:与模型组大鼠相比,西洋参虽不能显著改变肠道菌群的丰富度,却可显著改变菌群的多样性,门水平上主要表现为变形菌门比例降低(P<0.001),厚壁菌门和拟杆菌门升高(P<0.05或P<0.001),属水平上主要表现为假单胞菌属所占比例下降(P<0.05),拟杆菌属升高(P<0.001)。菌群功能分析显示西洋参可改善碳水化合物代谢、氨基酸代谢和能量代谢等。结论西洋参可以显著改善AAD大鼠腹泻程度,恢复结肠组织结构,其在改善AAD的同时可改善肠道菌群,恢复菌群碳水化合物等代谢过程。 Objective To study the intervention effect of Xiyangshen(Panacis Quinquefolii Radix)on antibiotic associated diarrhea(AAD)rats and analyze its effect on gut microbiota.Methods Antibiotic-associated diarrhea(AAD)model was established by giving antibiotic mixed solution by gavage.The control group(n=10)were treated with normal saline.The Xiyangshen(Panacis Quinquefolii Radix)groups(n=10)were treated separately with Xiyangshen(Panacis Quinquefolii Radix)decoction at 1 g/(kg·d)or 2 g/(kg·d).The diarrhea condition was evaluated.After treatment,the colon samples were collected and subjected to histological analysis.The changes of gut microbiota in rats were detected by high-throughput sequencing,and functional prediction was also carried out.Results AAD model rats showed obvious diarrhea symptoms.After the treatment of Xiyangshen(Panacis Quinquefolii Radix)decoction,compared with diarrhea rats,Xiyangshen(Panacis Quinquefolii Radix)could reduce the symptoms of diarrhea and improve AAD-related colon tissues histopathological symptoms.16 s rRNA sequencing analysis of gut microbiota showed that Xiyangshen(Panacis Quinquefolii Radix)could not significantly change the richness of intestinal flora,but it could significantly improve the diversity of intestinal flora composition compared with the model group.The level of phylum was mainly decreased by the proportion of Proteobacteria(P<0.001)and the increase of Bacteroidetes and Firmicutes(P<0.05 or P<0.001).The genus was mainly decreased by the proportion of Pseudomonas(P<0.05)and the increase of Bacteroides(P<0.001).Functional analysis showed that Xiyangshen(Panacis Quinquefolii Radix)could improve carbohydrate metabolism,amino acid metabolism and energy metabolism.Conclusion Xiyangshen(Panacis Quinquefolii Radix)can significantly improve the degree of diarrhea in AAD rats and restore the structure of colon tissue.While improving AAD,it can improve gut microbiota and restore carbohydrate and other metabolic processes of the flora.
