摘要
目的研究紫草素对小鼠乳腺癌4T1细胞增殖作用及其影响的相关机制。方法体外培养4T1细胞,使用不同浓度紫草素(0、12.5、25、50和100μmol/L)处理细胞,MTT分析其抑制细胞增殖的浓度;流式细胞术测定给药后对4T1细胞的周期与凋亡的影响;收集给药后的4T1细胞的蛋白,免疫印迹测定β-actin和MMP-2的表达和变化。结果紫草素能显著抑制小鼠乳腺癌细胞4T1的增殖,IC_(50)约为50μmol/L;一定浓度的紫草素能够阻滞细胞于G_(0)/G_(1)期。结论紫草素可显著抑制小鼠腺癌细胞增殖,且具有浓度依赖性。
Objective To study the mechanism of shikonin on proliferation and influence of mouse breast cancer 4T1 cells.Methods 4T1 cells were cultured in vitro and treated with different concentrations of shikonin(0,12.5,25,50 and 100μmol/L).The concentration of inhibiting cell proliferation was analyzed by MTT;Flow cytometry was used to determine the effects of administration on the cycle and apoptosis of 4T1 cells;The protein of 4T1 cells after administration was collected and determined by Western blotβ-Expression and changes of actin and MMP-2.Results Shikonin inhibited the proliferation of 4T1 cells,and the IC_(50) was about 50μmol/L.A certain concentration of shikonin can block cells in G_(0)/G_(1) phase.Conclusion Shikonin can significantly inhibit the proliferation of mouse adenocarcinoma cells in a concentration dependent manner.
