振动运动对2型糖尿病大鼠骨骼肌FOXO1、GLUT4蛋白表达的影响 被引量:2

Effects of Vibration Exercise on the Expression of FOXO1 and GLUT4 Protein in Skeletal Muscle of Type 2 Diabetic Rats

摘要 目的探讨振动运动对2型糖尿病大鼠糖脂代谢的影响及骨骼肌叉头框转录因子O亚族1(FOXO1)、丙酮酸脱氢酶激酶4(PDK4)、葡萄糖转运子4(GLUT4)蛋白表达的影响。方法2型糖尿病大鼠模型的制备,采用高脂高糖膳食喂养大鼠,腹腔注射链脲佐菌素(streptozotocin,STZ)的方法。采用高脂高糖饮食喂养雄性SD大鼠8 w后,腹腔注射链脲佐菌素(STZ)制备糖尿病大鼠模型,将造模成功的大鼠随机分组。运动方式:每天运动2次,每次15 min,振幅峰值为3 mm,每周运动6 d,共运动8 w。运动结束后,测定空腹血糖(fasting blood glucose,FBG)、血脂等生化指标;测定血清中IL-6、TNF-α含量,测定大鼠骨骼肌组织中超氧化物歧化酶(superoxide dismutase,SOD)的活性和丙二醛(malondialdehyde,MDA)含量。HE染色观察骨骼肌病理变化。Western blot检测FOXO1、PDK4、GLUT4蛋白的表达情况。结果振动运动中高频率组FBG、TG、TC、LDL-C含量与模型组比较显著降低,而HDL-C含量显著升高;血清中IL-6、TNF-α含量明显降低、骨骼肌中SOD的活性升高、MAD含量降低。HE染色显示中、高频率振动组大鼠骨骼肌断裂较少,肌丝走形紊乱明显改善。FOXO1、PDK4显著降低、GLUT4蛋白表达明显升高。结论振动运动可降低糖尿病大鼠血糖、血脂,降低FOXO1、PDK4蛋白表达、提高GLUT4蛋白表达,缓解糖尿病大鼠骨骼肌的病理损伤。 Objective To investigate the effects of vibration exercise on glucolipid metabolism and expression of FOXO1,Pyruvate dehydrogenase kinase 4(PDK4)and glucose transporter 4(GLUT4)in skeletal muscle of type 2 diabetic rats.Methods Type 2 diabetic rats were fed with high fat and high sugar diet and intraperitoneally injected with streptozotocin(STZ).Male SD rats were fed a high-fat and high-sugar diet for 8 w,then intraperitoneal injection of streptozotocin(STZ)was used to prepare diabetic rat model,and the successful model rats were randomly divided into two groups.Training method:Exercise twice a day with 15 min each time,the peak amplitude was 3 mm,and exercise for 6 days a week with a total of 8 weeks.After exercise,fasting blood glucose,blood lipid and other biochemical indicators were measured;the contents of IL-6 and TNF-αin serum,the activity of superoxide dismutase and the content of malondialdehyde in skeletal muscle of rats were determined.HE staining was used to observe the pathological changes of skeletal muscle.The expressions of FOXO1,PDK4 and GLUT4 proteins were detected by Western blot.Results The contents of FBG,TG,TC and LDL-C in the high frequency group were significantly lower than those in the model group,while the contents of HDL-C were significantly higher.The contents of IL-6 and TNF-αin serum decreased significantly,while SOD activity increased and MAD content decreased in skeletal muscle.HE staining showed that skeletal muscle fracture was less and myofilaments dislocation disorder was improved significantly in rats of medium and high frequency vibration group.FOXO1 and PDK4 decreased significantly,and GLUT4 protein expression increased significantly.Conclusion Vibration exercise can decrease blood glucose and lipid,decrease FOXO1 and PDK4 protein expression,increase GLUT4 protein expression,and alleviate pathological injury of skeletal muscle in diabetic rats.
