摘要
目的:建立中药三七的 LC-MS化学模式识别方法,用于三七不同药用部位(绒根、剪口、根条和主根)的区分,为其质量评价提供可靠数据。方法:应用 LC-MS技术建立中药三七不同部位的指纹图谱,获取三七不同部位间的峰面积信息,将指纹图谱与化学模式识别的方法相结合,利用无监督的聚类分析(HCA)、主成分分析(PCA)和有监督方法偏最小二乘法判别分析(PLS-DA)对其进行数据处理,建立质量分类模型,寻找三七不同部位间的质量差异,以期建立可以区分中药三七不同部位的质量分类模型。结果:采用 PLS-DA 法经过特征提取,筛选出4个特征性指标,四类样品能够实现准确的区分,经过与标准品比对,分别鉴定为人参皂苷 Rb_(1)、人参皂苷 Rb_(2)、人参皂苷 Rb_(3)和20(S)-人参皂苷-Rg_(2);通过内部验证 ,其 R^(2)和 Q^(2)均符合要求,外部验证的准确率为100%,确证了分类模型的可靠性和准确性。结论:得到的4个特征性成分可作为典型质量标记物用于区分三七不同药用部位,从而对三七质量评价提供数据支持。
Objective:The study aims to establish a method of chemical pattern recognition method coupled with LC-MS for the quality evaluation of Panax notoginseng (Burk.)F.H.Chen (P.notoginseng)including different medicinal parts(downy roots,cuts,root strips and tap roots)in order to provide a reliable data support for its quality.Methods:LC-MS fingerprints of different parts of P.notoginseng were obtained concerning their peak area.The method of LC-MS fingerprints and chemical pattern recognition including unsupervised hierarchical cluster analysis(HCA),principal component analysis(PCA)and supervised partial least squares discriminant analysis(PLS-DA)were combined to establish a quality classification model thus find quality marks for distinguishing the difference of different parts of P.notoginseng.Results:PLS-DA method was proved the effective model for their quality evaluation.Hopefully,four characteristic peaks were selected and all the selected samples could be accurately distinguished.In addition,those peaks were identified with reference materials as Ginsenoside Rb_(1),Ginsenoside Rb_(2),Ginsenoside Rb_(3) and 20(S)-Ginsenoside-Rg_(2).Moreover,the established PLS-DA model was validated by internal verification(R^(2) and Q^(2))and external verification.It was found the results were all satisfactory.Conclusion:The selected four characteristic peaks could be used as potential quality markers to distinguish different parts of P.notoginsengand to provide data support for the quality evaluation of P.notoginsengas well.
