摘要
为研究石莲花多糖的复合酶法提取工艺及其护肤特性,将新鲜的石莲花多肉去皮以纤维素酶和果胶酶组成的复合酶分解提取多糖,以多糖提取率为评价指标,优化提取工艺,并通过皮肤水分测试、风光光度计和羟自由基的清除率来评价其保湿性、抗紫外活性和抗氧化性。可知石莲花多糖的最佳工艺条件为复合酶用量为5.0%、料液比1∶20(g/mL)、酶解温度为50℃、酶解时间为270 min、pH值为3,在此条件下,提取得石莲花粗多糖的含量大致在6.09%左右皮肤水分测试在1 h内数值较好,在紫外波段有峰值并且其羟自由基清除率为51.92%。因此,复合酶法提取操作简单,优化后能够提高提取率,石莲花多糖具有一定的保湿性、抗紫外活性和抗氧化性。
Objective:To study the compound enzymatic extraction process and skin care properties of horseleek polysaccharide.Methods:The fresh horseleek succulent was peeled and the polysaccharide was decomposed and extracted by the compound enzyme composed of cellulase and pectinase.The extraction rate of polysaccharide was used as the evaluation index,and the extraction process was optimized.The moisture retention,anti UV activity and antioxidant activity were evaluated by skin moisture test,spectrophotometer and hydroxyl radical clearance rate.Results:The optimum process conditions of horseleek polysaccharide were as follows:compound enzyme dosage was 5.0%,solid-liquid ratio was 1:20(g/ml),enzymatic hydrolysis temperature was 50℃,enzymatic hydrolysis time was 270 min,and pH value was 3.Under these conditions,the content of horseleek crude polysaccharide was about 6.09%,the value of skin moisture test was good within 1h,there was a peak in UV band,and its hydroxyl radical scavenging rate was 51.92%.Conclusion:The compound enzymatic extraction was simple,and the extraction rate could be improved after optimization.The polysaccharide of horseleek had certain moisturizing,anti-ultraviolet activity and antioxidant properties.
