摘要
目的研究灵芝总三萜(GLTs)对对乙酰氨基酚(APAP)诱导急性肝损伤的保护作用。方法48只Wistar大鼠随机分成6组,分别为空白对照组、APAP肝损伤模型对照组、GLTs低剂量组50 mg/(kg·bw)、中剂量组100 mg/(kg·bw)和高剂量组200 mg/(kg·bw)和N-乙酰半胱氨酸(NAC,200 mg/(kg·bw))阳性对照组。测定血清的谷氨酸氨基转移酶(ALT)、天冬氨基转移酶(AST)和乳酸脱氢酶(LDH)含量,肝组织的丙二醛(MDA)、谷胱甘肽(GSH)和超氧化物歧化酶(SOD)水平,H&E染色观察肝组织病理学变化,Western-blotti ng测定肝组织HO-1蛋白的表达情况。结果与模型组比较,GLTs高剂量组均能降低血清中ALT、AST和LDH水平(P<0.05),降低肝匀浆MDA水平,提高肝匀浆GSH和SOD含量,HO-1蛋白的表达增加。灵芝总三萜各剂量组对APAP引起的肝损伤均有一定程度改善作用。结论灵芝总三萜对APAP诱导的肝损伤具有明显的保护作用,具有开发成保健品或药品的潜力。
OBJECTIVE The protective effect of total triterpenoids from Ganoderma lucidum(Leyss.ex Fr.)Karst.(GLTs)on acetaminophen(APAP)-induced liver injury in rats was investigated.METHODS 48 Wistar rats we re randomly divided into 6 groups,including blank control group,APAP-induced liver injury model group,and low-dose(50 mg/(kg·bw)),middle-dose(100 mg/(kg·bw)),and high-dose(200 mg/(kg·bw))GLTs treated groups,as well as positive control group(NAC,200 mg/(kg·bw)).The serum contents of ALT,AST and LDH,and the hepatic content of MDA,GSH,and SOD activities were determined.The pathological changes of liver tissue were observed by H&E staining.The expression of HO-1 protein in liver was detected by Western-blotting.RESULTS The results showed that high dose GLTs markedly decreased the content of ALT,AST and LDH in serum,and MDA in liver tissue.Meanwhile,hepatic GSH content and SOD activity,HO-1 protein expression were significantly increased in high dose GLTs group,compared with APAP-induced liver injury model group.Different doses of GLTs could ameliorate APAP-induced liver injure in certain degree.CONCLUSION GLTs presented significant protective effect on APAP-induced liver injury,thus presenting the potential to be developed into health care products or drugs.
