食用菌中无机汞和甲基汞的分析方法研究 被引量:2

摘要 目的建立液相色谱-原子荧光光谱联用技术测定食用菌中无机汞和甲基汞含量的方法。方法样品经盐酸溶液超声提取,氨水中和,L-半胱氨酸溶液衍生,采用C;色谱柱,以5%甲醇-0.03 mol/L乙酸铵-0.1%L-半胱氨酸为流动相进行分离,经原子荧光光谱进行检测。结果当样品称样量为0.5 g,定容体积为25 mL时,无机汞和甲基汞的检出限分别为0.002 mg/kg和0.001 mg/kg,方法线性范围为0~50.0 ng/mL,线性相关系数R^(2)=0.9999,不同浓度水平精密度良好(RSD<3.0%),三个不同浓度水平的加标回收率范围为79.0%~104.3%。结论本方法操作简便、灵敏度高,适用于市售食用菌中无机汞和甲基汞的测定。通过对市售食用菌分析研究,发现汞含量高的食用菌主要以无机汞形式存在,甲基汞含量较低。 Objective To develop a method for the determination of inorganic mercury and methylmercury in edible mushrooms by liquid chromatography-atomic fluorescence spectrometry(LC-AFS).Methods The samples were ultrasonically extracted with hydrochloric acid solution,neutralized with ammonia and derivatized with L-cysteine solution,separated on a C;analytical column using 5%methanol-0.03 mol/L ammonium acetate-0.1%L-cysteine as the mobile phase,and detected by atomic fluorescence spectroscopy.Results When the sample was weighted at 0.5 g and the volume was fixed 25 mL,the limits of detection of inorganic mercury and methylmercury were 0.002 mg/kg and 0.001 mg/kg,respectively.The linearity range of the method was 0-50.0 ng/mL,and the linear correlation coefficient was R^(2)=0.9999 with good precision(RSD<3.0%).The recoveries of standard addition for different matrices at three different concentration levels ranged from79.4%to 104.3%.Conclusions The method is simple,sensitive and suitable for the determination of inorganic mercury and methylmercury in commercially available edible mushrooms.Through the analysis of commercially available edible mushrooms,it is found that edible mushrooms with high mercury content are mainly in the form of inorganic mercury,and low in methylmercury.
