菟丝子不同炮制品指纹图谱及多指标成分定量研究 被引量:2

摘要 目的建立菟丝子指纹图谱及多指标含量测定方法探究菟丝子炮制后的变化。方法以Agilent SB柱(100 mm×2.1 mm,1.8μm)为色谱柱,乙腈(A)-0.1%磷酸(B)为流动相,流速为0.3 mL·min^(-1),柱温为40℃,检测波长为360 nm。采用相似度评价结合主成分分析、正交偏最小二乘法-判别分析等模式探讨影响菟丝子炮制前后指纹图谱中总体化学成分的变化。结果菟丝子生品和炮制品指纹图谱具有一定的差异,菟丝子标定了10个共有峰,炒菟丝子和盐菟丝子标定了12个共有峰,其中峰11、峰12为炒菟丝子和盐菟丝子炮制后产生的新成分,分别为新绿原酸、隐绿原酸。指认了8个色谱峰并对其进行定量分析,结果显示,新绿原酸、绿原酸、隐绿原酸、金丝桃苷和槲皮素含量在炮制后发生了变化。结论该方法可控制菟丝子炮制品的质量。 Objective To determine the change of Cuscuta australis R.Br.before and after being processing by fingerprint and multi-index determination method.Methods The column was Agilent SB(100 mm×2.1 mm,1.8μm)and acetonitrile-0.1%phosphoric acid was used as the mobile phase with gradient elution.The flow rate was 0.3 mL·min^(-1).The column temperature was 40℃.The detection wavelength was 360 nm.Similarity evaluation,principal component analysis and orthogonal partial least squares-discriminant analysis were conducted to explore the changes in total chemical ingredients of Cuscuta australis R.Br.before and after the processing.Results There were some differences in the fingerprints of Cuscuta australis R.Br.and its processed products.Totally 10 common peaks were marked in Cuscuta australis R.Br.,while 12 common peaks were marked in fried-processed Cuscuta australis R.Br.and salt-processed Cuscuta australis R.Br..Peak 11 and 12(neochlorogenic acid and cryptochlorogenic acid)were the new components generated after the processing.Totally 8 peaks were identified and measured.Analysis of variance displayed the changes of neochlorogenic acid,chlorogenic acid,cryptochlorogenic acid,hyperoside and quercetin after the processing.Conclusion This method can control the quality of processed Cuscuta australis R.Br..
