基于聚氨酯海绵载体的棘孢木霉固体发酵培养基筛选 被引量:3

Screening of Solid Fermentation Medium for Trichoderma asperellum Based on Polyurethane Sponge Carrier

摘要 以棘孢木霉Trichoderma asperellum THGY-01(CGMCC No:22422)为试验菌株,对其在聚氨酯海绵载体上固体发酵产孢的培养基种类、浓度及促进产孢元素进行了筛选与优化。以聚氨酯海绵载体的单位质量产孢量为指标,通过单因素试验法对11种碳源、6种氮源和14种无机盐(8种含大量元素,6种含微量元素)进行了筛选。结果表明:糖蜜为产孢最佳的碳源,发酵6 d时,浓度为5%(V/V)处理的产孢量达4.92×10^(9)孢子/g;发酵8 d时,浓度为30.0%(V/V)处理的产孢量达到18.86×10^(9)孢子/g,其产孢量是其他碳源最高产孢量的11.45倍;筛选出产孢最佳的氮源为酵母粉,发酵6d时,浓度为0.4%(M/V)处理的产孢量可达1.42×10^(9)孢子/g,其产孢量是其他氮源最高产孢量的1.27倍。研究还发现,一定浓度范围内含钾离子的无机盐可提高聚氨酯海绵载体上木霉菌固体发酵的孢子产量,向培养基中添加浓度为0.20%(M/V)的KH_(2)PO_(4),发酵6 d时孢子产量达2.31×10^(9)孢子/g,比对照产孢量高203倍;一些微量元素如Cu、Zn、Mn在一定浓度范围内对菌株的产孢有显著的促进作用。该研究为棘孢木霉在聚氨酯海绵载体上的固体发酵提供了理论基础。 Using Trichoderma asperellum THGY-01(CGMCC No: 22422) as the test strain, we screened and optimized the type and concentration of spore-producing medium for solid fermentation on a polyurethane sponge carrier, and the elements that could promote sporulation were also identified. Using the spore production per unit mass of the polyurethane sponge carrier(spores/g) as an indicator, 11 types of carbon sources, 6 types of nitrogen sources and 14 types of inorganic salts(8 types containing large elements and 6 types containing trace elements)were screened by the single factor test method. The results showed that molasses was the best carbon source for sporulation;When fermentation with 5%(V/V) molasses for 6 days, the spore production reached 4.92×10^(9)spores/g;When fermentation with 30.0%(V/V) for 8 days, the spore production even reached 18.86×10^(9)spores/g,which was 11.45 times the highest spore production of other carbon sources. The best nitrogen source for sporulation was yeast powder. After 6 days of fermentation with 0.4%(W/V) yeast powder, the spore production reached 1.42×10^(9)spores/g, which is 1.27 times the highest spore production of other nitrogen sources. We also found that adding a certain concentration of potassium ion-containing inorganic salt can increase the spore yield of Trichoderma solid fermentation on the polyurethane sponge carrier;with the addition of 0.20%(M/V) KH_(2)PO_(4), the spore production reached 2.31×10^(9)spores/g after 6 days of fermentation, which was 203 times higher than the control;Some trace elements such as Cu, Zn, and Mn at certain concentration also significantly promote the sporulation of tested strains. The above results provide a theoretical basis for the solid fermentation of Trichoderma asperellum on a polyurethane sponge carrier.
