绵羊HERC1和ALDH6A1多态性与胴体性状的关联分析

摘要 本研究旨在探究绵羊HERC1与ALDH6A1多态性与胸椎数和胴体体长间的相关性,筛选提高绵羊胴体品质的重要分子标记。利用Sequenom MassARRAY SNP技术检测苏尼特羊和小尾寒羊两个绵羊群体与胴体性状相关候选基因HERC1和ALDH6A1单核苷酸多态性(SNPs)位点,进行群体遗传学分析,并与表型性状关联;利用生物信息学工具对HERC1分子结构进行预测分析。结果表明,HERC1 c.8950G>A、g.43407058T>G、c.11902A>G和ALDH6A1 c.1126A>G在小尾寒羊和苏尼特羊中均与胸椎数不显著相关。SNPs与胴体体长分析结果表明,在小尾寒羊HERC1 c.8950G>A中,野生型(GG)胴体体长显著高于突变型(GA)(P<0.05)。生物信息学分析表明,HERC1为亲水蛋白,蛋白结构不稳定,突变前后均无跨膜结构域,可能不参与跨膜间生化反应。综上所述,HERC1 c.8950G>A位点与小尾寒羊胴体体长显著相关,可作为提高绵羊胴体性状的潜在分子辅助标记。 The aim of this study was to investigate the correlation between HERC1 and ALDH6A1 polymorphisms and the number of the thoracic vertebrae and carcass length,and to screen important molecular markers to improve carcass quality for sheep.Detection of candidate genes HERC1 and ALDH6A1 single nucleotide polymorphisms(SNPs)loci associated with carcass traits in two sheep populations,Sunit and Little Tail sheep,using Sequenom MassARRAY SNP technology for population genetics analysis and association with phenotypic traits.Predictive analysis of HERC1 molecular structure using Bioinformatics tools.The results showed that HERC1 c.8950G>A,g.43407058T>G,c.11902A>G and ALDH6A1 c.1126A>G were not significantly correlated with the number of thoracic vertebrae in both Small Tail Han sheep and Sunit sheep.The results of SNPs and carcass length analysis showed that the carcass length was significantly higher(P<0.05)in the wild type(GG)than in the mutant type(GA)in the Small Tail Han sheep HERC1 c.8950G>A.Bioinformatics analysis showed that HERC1 is a hydrophilic protein with an unstable protein structure and no transmembrane structural domain before and after the mutation,which may not participate in transmembrane inter-biochemical reactions.In conclusion,the HERC1 c.8950G>A locus was significantly associated with carcass length in Small Tail Han sheep and could be used as a potential molecular auxiliary marker to improve carcass traits for sheep.
