肿瘤相关巨噬细胞、胶质瘤干细胞和PF-L1在不同病理分级脑胶质瘤组织中的表达及相关性研究 被引量:2

摘要 目的 探讨人脑胶质瘤组织内的胶质瘤干细胞(GSCs)标志物CDl33、肿瘤相关巨噬细胞(TAMs)标志物CD68和PD-L1在不同病理分级脑胶质瘤组织中表达及其相关性。方法 选取脑胶质瘤患者68例,根据病理分级不同,分为低级别组34例(WHOⅢ级、Ⅳ级),高级别组34例(WHOⅠ级、Ⅱ级),取两组标本进行石蜡包埋切片,应用免疫荧光法检测不同病理分级脑胶质瘤组织内CD68和PD-L1蛋白的含量;采用实时荧光定量PCR技术检测两组脑胶质瘤组织内的CDl33、PD-L1和CD68 mRNA的表达,并分析其与临床病理级别的相关性。结果 在高级别组脑胶质瘤组织中CD133、CD68和PD-L1表达量较低级别组升高,差异显著(P<0.001)。在脑胶质瘤组织内CD133、PD-L1和CD68的表达与不同病理分级呈正相关(P<0.001)。结论 PD-L1在胶质瘤组织内主要通过肿瘤微环境中的TAMs进行表达;脑胶质瘤的恶性程度与CDl33、PD-L1和CD68表达密切相关。 Objective To investigate the expression and correlation of CD133,CD68 and PD-L1 in different pathological grades of human glioma.Methods 68 patients with gliomas were selected and divided into the low-grade group(34 cases,who grade Ⅲ and Ⅳ) and high-grade group(34 cases,who grade Ⅰ and Ⅱ) according to different pathological grades.The samples of the two groups were embedded in paraffin,and the contents of CD68 and PD-L1 protein in gliomas of different pathological grades were detected by immunofluorescence method;The expressions of CDl33,PD-L1 and CD68 mRNA in glioma tissues of the 2 groups were detected by real-time fluorescence quantitative PCR,and their correlation with clinicopathological grade was analyzed.Results The expressions of CD133,CD68 and PD-L1 were significantly higher in high-grade group than those in low-grade group(P<0.001).The expressions of CD133,PD-L1 and CD68 in glioma were positively correlated with different pathological grades(P<0.001).Conclusion PD-L1 is expressed in glioma mainly through TAMs in tumor microenvironment;The malignant degree of glioma is closely related to the expression of CDl33,PD-L1 and CD68.
