摘要
本研究基于前期的试验结果,以花育6303为研究对象,克隆了花生耐盐转录因子AhbHLH18。通过花序侵染法将AhbHLH18转录因子转化拟南芥,并通过卡那霉素筛选和PCR鉴定,获得转基因拟南芥。盐胁迫处理后,相对于野生型拟南芥,转基因拟南芥具有更显著的耐盐表型、更高的抗氧化酶活性和更低的MDA含量。本研究结果可为进一步利用该转录因子提供理论依据。
Based on our previous research, in this study, we characterized the function of a transcription factor bHLH as AhbHLH18 in the peanut from Huayu6303. The AhbHLH18 was transformed into Arabidopsis thaliana by the floral dip method, and then, the transgenic plant was screened using Kanamycin resistance and PCR identification. Under salt treatment, compared to wild-type plant, antioxidant enzymes(superoxide dismutase, peroxidaseand catalase) in the transgenic plants were more active. In addition, the wild-type plant had higher MDA concentration level than the transgenic plant. This study provided a basis for utilization of this transcription factor.
