摘要
利用G-四链体DNA(T30695)催化Zn^(2+)插入到中卟啉IX(MPIX)中,引起荧光偏移的特点,建立了检测Zn^(2+)的方法。在40μmol/L MPIX、0.6μmol/L Pb^(2+)、5μmol/L T30695和1%Triton的最优实验条件下,该方法在Zn^(2+)浓度为0.5~5μmol/L范围内呈现良好的线性关系,相关系数R^(2)=0.95,检出限为73.5 nmol/L。离子选择性实验表明该方法对Zn^(2+)具有较好的选择性,用于实际样品测定,回收率在94.7%~100.4%之间。
A new biosensor was developed to detect zinc ion based on the principle that G-quadruplex DNA(T30695)catalyzed the insertion of zinc ion into mesoporphyrin IX(MPIX)and caused a shift in fluorescence spectra.Under optimal experimental conditions(40 μmol/L MPIX,0.6 μmol/L Pb^(2+),5 μmol/L T30695 and 1% Triton),this method had a good linear relationship in the range of 0.5-5 μmol/L(R~2=0.95),with a detection limit of 73.5 nmol/L.The selectivity experiments showed that this method had a good selectivity for zinc ion.Satisfactory recovery rates(94.7%-100.4%) were obtained,indicating that this method could be used in practical applications.
