摘要
目的 探讨T2DM大鼠环状RNA(circRNA)表达与IR及胰岛β细胞功能的相关性。方法 选取SPF级SD大鼠18只,随机选取6只为正常对照(NC)组,其余12只为T2DM组。测量体重,检测FPG、TC、TG、BUN、血肌酐(Scr),行腹腔注射糖耐量试验和胰岛素耐量试验,计算胰岛素抵抗指数(HOMA-IR)、胰岛β细胞功能指数(HOMA-β)及胰岛素敏感指数(HOMA-IS),HE染色,计算胰岛个数并测量胰岛面积,荧光定量PCR检测rno_circRNA_0003340基因表达量。结果 与NC组比较,T2DM组FPG、TC、TG、HbA1c、BUN、FIns、HOMA-IR、rno_circRNA_0003340表达水平升高(P<0.05或P<0.01),体重、Scr、HOMA-β及HOMA-IS降低(P<0.01)。T2DM组胰岛个数和胰岛面积均低于NC组(P<0.05)。偏相关分析显示,T2DM组外周血单个核细胞rno_circRNA_0003340表达水平与Flns、HOMA-IR呈正相关(P<0.05),与HOMA-β、HOMA-IS呈负相关(P<0.05)。结论rno_circRNA_0003340表达水平可能与T2DM大鼠IR、胰岛β细胞功能密切相关,rno_circRNA_0003340表达水平上调可能参与T2DM的发生发展。
Objective To investigate the expression of circRNA in rats with type 2 diabetes mellitus(T2DM)and the relationship between circRNA and insulin resistance(IR)and islet β-cell function.Methods Eighteen SPF SD rats were selected.Six rats were randomly selected as normal control(NC)group and the other 12 rats were selected as T2DM group.Body weight was measured.FPG,TC,TG,urea nitrogen(BUN),serum creatinine(Scr)were tested.Intraperitoneal injection glucose tolerance test(IPGTT)and insulin tolerance test(ITT)were performed.HOMA-IR,HOMA-β and insulin sensitivity index(HOMA-IS)were calculated in all the subjects.The number of islet cells and islet area were evaluated by HE staining.Results FPG,TC,TG,HbA;c,BUN,FIns,HOMA-IR and expression level of rno_circRNA_0003340were higher in T2DM group than in NC group(P<0.05 or P<0.01).Body weight,Scr,HOMA-β and HOMA-IS were lower in T2DM group than in NC group(P<0.01).The number and area of pancreatic islets were lower in T2DM group than in NC group(P<0.05).Partial correlation analysis showed that the expression level of rno_circRNA_0003340 gene in peripheral blood mononuclear cells was positively correlated with Flns and HOMA-IR(P<0.05),and negatively correlated with HOMA-β and HOMA-IS(P<0.05)in T2DM group.Conclusion The expression level of rno_circRNA_0003340 may be closely related to IR and pancreatic islet β -cell function in type 2 diabetic rats.The expression level of rno_circRNA_0003340up-regulation may be involved in the development of T2DM.
