摘要
目的分析舒肝解郁胶囊对抑郁症大鼠的改善作用及对海马神经元细胞的修复作用。方法50只SD雄性大鼠随机分为对照组、模型组、舒肝解郁胶囊组、阳性药物组和舒肝解郁胶囊+AG490(JAK激酶抑制剂)组。用敞箱实验和糖水消耗实验评价大鼠行为;用Nissl与Tunnel染色观察海马神经元细胞存活与凋亡情况;用实时荧光定量PCR、Western blot检测海马组织酪氨酸蛋白激酶-1(tyrosine-protein kinase 1,JAK1)、信号转导激活转录因子-3(signal transducer activated transcription factor-3,STAT3)mRNA和蛋白的表达情况。结果与对照组比较,模型组敞箱实验得分、糖水消耗百分比与神经元细胞存活数量降低,凋亡神经元数量增加(P<0.01),海马组织磷酸化JAK1(p-JAK1)/JAK1、磷酸化STAT3(p-STAT3)/STAT3降低(P<0.01);与模型组比较,阳性药物组敞箱实验得分、糖水消耗百分比和神经元细胞存活数量增加,神经元细胞凋亡数量减少(P<0.01),海马组织p-JAK1/JAK1、p-STAT3/STAT3升高(P<0.01);与阳性药物组比较,舒肝解郁胶囊组敞箱实验得分、糖水消耗百分比和神经元细胞存活数量减少,神经元细胞凋亡数量增加(P<0.01),海马组织p-JAK1/JAK1、p-STAT3/STAT3降低(P<0.01);与舒肝解郁胶囊组比较,舒肝解郁胶囊+AG490组敞箱实验得分、糖水消耗百分比和神经元细胞存活数量减少,神经元细胞凋亡数量增加(P<0.01),海马组织p-JAK1/JAK1、p-STAT3/STAT3降低(P<0.01)。结论舒肝解郁胶囊可改善抑郁症大鼠的行为,修复损伤的海马神经元细胞,其可能通过激活JAK1/STAT3信号通路相关基因和蛋白的表达发挥作用。
Objective To analyze the improvement effect of Shugan Jieyu Capsules on depression rats and the repair ment effect on hippocampal nerve cells.Methods 50 male SD rats were randomly divided into control group,model group,Shuganjieyu Capsules group,positive drug group,Shuganjieyu Capsules+AG490(JAK kinase inhibitor)group.The rats’behavior was evaluated by open field test and sugar water consumption test.Nissl and tunnel staining were used to observe the survival and apoptosis of hippocampal neurons.The mRNA and protein expressions of hippocampal neuron damage through tyrosine protein kinase-1(JAK1)and ignal transducer and activator of transcription 3(STAT3)were detected by real-time fluorescence quantitative PCR and Western blot.Results Compared with the control group,the open box test score,the percentage of sugar and water consumption and the survival number of neuronal cells in the model group decreased,the number of neuronal apoptosis increased(P<0.01),and the phosphorylated JAK1(p-JAK1)/JAK1 and phosphorylated STAT3(p-STAT3)/STAT3 in hippocampus decreased(P<0.01).Compared with the model group,the score of open box experiment,the percentage of sugar and water consumption and the survival number of neuronal cells in the positive drug group increased,the number of neuronal apoptosis was decreased(P<0.01),while the p-JAK1/JAK1 and p-STAT3/STAT3 in hippocampus increased(P<0.01).Compared with the positive drug group,the open box test score,the percentage of sugar and water consumption and the survival number of neuronal cells in Shugan Jieyu Capsules group decreased,the number of neuronal apoptosis increased(P<0.01),and the p-JAK1/JAK1 and p-STAT3/STAT3 in hippocampus decreased(P<0.01).Compared with Shugan Jieyu Capsules group,the Shugan Jieyu Capsules+AG490 group had lower open box test score,percentage of sugar and water consumption and the survival number of neuronal cells,while the number of apoptotic neurons increased(P<0.01),and hippocampal tissue p-JAK1/JAK1,p-STAT3/STAT3 decreased(P<0.01).Conclusion Shuganjieyu Capsules can improve the behavior of depression rats and repair the damaged hippocampal neurons,and the mechanism may play a role by activating the expression of JAK1/STAT3 signaling pathway related genes and proteins.
