孔雀源蛔虫分子鉴定及其遗传进化分析

摘要 本研究的主要目的是通过分析遗传变异探讨孔雀源蛔虫与其他蛔虫种群的遗传关系。本研究中,以湖南省益阳市楠蓝孔雀生态养殖场的8只孔雀小肠中采集蛔虫样品为研究对象,通过PCR方法对其线粒体cox1基因部分(pcox1)序列和nad1基因部分(pnad1)序列以及核糖体ITS序列进行扩增、测序和分析,并利用邻接法(Neighbor joining)基于3个基因位点构建孔雀源蛔虫和其它蛔虫的进化树,分析其种系发育关系。测序结果显示,8只孔雀源蛔虫样品的pcox1、pnad1和ITS基因序列长度分别为442、512、1023~1029 bp,碱基差异分别为0.00%~0.45%、0.00%~0.39%和0.00%~0.87%,系统进化分析均显示8只孔雀源蛔虫与已知鸡蛔虫分离株聚类形成同一分支,与其他蛔虫所属分支分隔较远,说明本研究获得的8只孔雀源蛔虫均被鉴定为鸡禽蛔虫(Ascaridia galli),其研究结果可为孔雀蛔虫病的防控提供指导。 The aim of the present study was to explore the population genetic relationships between ascarid of peacock origin and other species by the analysis of the genetic variation.In this research,ascarid was collected from the small intestines of 8 peafowls from Nanlan farm in Yiyang,Hunan province.The partial sequence of mitochondrial cox1(pcox1)and nad1(pnad1)as well as the full sequence of ribosomal ITS gene were amplified by PCR.Then,the Neighbor-Joining phylogenetic trees of peacock-derived ascarid and other ascarid were constructed based on three gene loci to analyze their phylogenetic relationships.The sequencing results showed that the lengths of pcox1,pnad1 and ITS genes of 8 samples were 442,512,1023-1029 bp,and the base differences were 0.00%-0.45%,0.00%-0.39%,and 0.00%-0.87%,respectively.Further phylogenetic analysis revealed that all 8 samples formed a sub-cluster with known Ascaris galli isolates,which was far apart from the branches of other ascarid.Thus,all 8 samples obtained in this study were identified as Ascaris galli.This study contributed useful information to the prevention and control of the ascarid disease.
