Discussion on the Mechanism of Preventing and Treating Atherosclerosis with the Modified Baohe Pills Based on Network Pharmacology

摘要 目的基于网络药理学探讨加减保和丸防治动脉粥样硬化(AS)的作用机制。方法通过中药系统药理学数据库与分析平台(TCMSP)及SwissTargetPrediction数据库等获取加减保和丸活性成分及靶点,利用UniProt数据库规范靶点基因名称;借助GeneCards、OMIM、TTD、DrugBank、PharmGKB数据库筛选AS疾病靶点,通过STRING数据库及Cytoscape3.8.2软件构建蛋白相互作用网络,并对交集靶点进行GO功能与KEGG通路富集分析。结果共筛选出加减保和丸防治AS的活性成分189个及其对应的靶点768个,核心靶点45个。其中CCL2、IL-6、TNF、SERPINE1等与AS的关键靶点相关联,参与炎症反应、细胞增殖、类固醇代谢等生物过程,作用于癌症中的蛋白多糖、类风湿关节炎、乙型肝炎、疟疾及HIF-1、Rap1、IL-17等信号通路。结论加减保和丸主要成分槲皮素、β-谷甾醇和山柰酚,可能参与调控关键靶点CCL2、IL-6、TNF、SERPINE1,抑制脂多糖和类固醇代谢,调节免疫和炎症反应,保护血管内皮细胞,以防治AS。 Objective To discover the mechanism of modified Baohe Pills in preventing and treating atherosclerosis(AS)based on network pharmacology.Methods TCMSP and SwissTargetPrediction database were used to obtain the active components and targets in modified Baohe Pills;AS disease targets were screened with the help of GeneCards,OMIM,TTD,DrugBank,PharmGKB databases.Protein-protein interaction network was constructed by STRING database and Cytoscape3.8.2 software.GO function and KEGG pathway enrichment analysis were conducted for the intersection targets.Results Totally 189 active components contained in the modified Baohe Pills and their corresponding 768 targets and 45 core targets were screened out.Among them,CCL2,IL-6,TNF,SERPINE1,etc.were associated with key targets of AS,involved in biological processes such as inflammation response,cell proliferation,and steroid metabolism,and were involved in proteoglycans in cancer,rheumatoid arthritis,hepatitis B,malaria diseases,and signaling pathways such as HIF-1,Rap1 and IL-17.Conclusion Quercetin,β-sitosterol and kaempferol are the main components of modified Baohe Pills,which may be involved in regulating the key targets of CCL2,IL-6,TNF,SERPINE1,inhibiting lipopolysaccharide and steroid metabolism,regulating immune and inflammatory responses,and protecting vascular endothelial cells,in order to prevent AS.
