摘要
目的 探究丹酚酸B对肺炎链球菌肺炎幼鼠的保护作用和机制。方法 90只幼鼠分为对照组、模型组、丹酚酸B低、中、高剂量组,除对照组外其余建立肺炎链球菌肺炎模型,丹酚酸B低、中、高剂量组分别灌胃10、20、40 mg/kg的丹酚酸B,每日1次,连续7 d,对照组和模型组灌胃等量生理盐水。肺功能仪检测用力肺活量(FVC)和呼气峰流速(PEF);HE染色观察肺组织病理学变化;ELISA法检测肺组织肿瘤坏死因子(TNF-α)和白细胞介素-6(IL-6)水平;RT-qPCR检测肺组织髓样分化因子88(MyD88)、核因子κB p65(NF-κB p65)和Nod样受体蛋白3(NLRP3)mRNA表达水平;Western blot检测肺组织MyD88、NF-κB p65、p-NF-κB p65和NLRP3蛋白表达水平。结果 模型组幼鼠肺组织结构模糊,有大量炎性细胞浸润,丹酚酸B各剂量组幼鼠炎性浸润减轻;与模型组比较,丹酚酸B各剂量组幼鼠FVC和PEF升高,TNF-α和IL-6水平、MyD885、NLRP3 mRNA和蛋白表达水平以及p-NF-κB p65蛋白表达水平降低,且上述改变有剂量依赖性(P<0.05)。结论 丹酚酸B对肺炎链球菌肺炎幼鼠具有治疗作用,其机制可能与抑制MyD88/NF-κB/NLRP3通路,减轻炎症反应有关。
Objective To investigate the protective effect and mechanism of salvianolic acid B on young mice with Streptococcus pneumoniae pneumonia. Methods Ninety young mice were divided into control group, model group, salvianolic acid B low-dose, medium-dose and high-dose groups. Except for the control group, the others were established to establish Streptococcus pneumoniae pneumonia model. Salvianolic acid B at doses of 10, 20,and 40 mg/kg, once a day, for 7 consecutive days, the control group and model group were given the same amount of normal saline by gavage. After intervention, forced vital capacity(FVC) and peak expiratory flow rate(PEF)were measured by pulmonary function instrument. HE staining was used to observe the pathological changes of lung tissue. Tumor necrosis factor(TNF-α) and interleukin-6(IL-6) levels in lung tissue were determined by ELISA. The mRNA expression levels of myeloid differentiation factor 88(MyD88), nuclear factor κB p65(NF-κB p65) and Nod-like receptor protein 3(NLRP3) in lung tissues were detected by RT-qPCR. The protein expression levels of MyD88, NF-κB p65, p-NF-κB p65 and NLRP3 in lung tissues were detected by Western blot. Results The lung tissue structure of the young mice in the model group was blurred, with a large number of inflammatory cells infiltrated. The inflammatory infiltration of the young mice in the salvianolic acid B dose groups was alleviated. Compared with the model group, the FVC and PEF of the young mice in the salvianolic acid B dose groups increased, TNF-α and IL-6 levels, MyD885, NLRP3 mRNA and protein expression levels, and p-NF-κB p65 protein expression levels were decreased in a dose-dependent manner(P < 0.05). Conclusion Salvianolic acid B has therapeutic effect on young rats with Streptococcus pneumoniae pneumonia, and its mechanism may be related to inhibiting MyD88/NF-κB/NLRP3 pathway and alleviating inflammatory response.
