摘要
目的探讨湿润烧伤膏对糖尿病皮肤溃疡小鼠肌肉组织中核因子NF-E2相关因子(nuclear factor E2-related factor 2,Nrf2)、醌氧化还原酶-1(nadph:quinone oxidoreductase-1,NQO1)、血红素加氧酶-1(heme oxygenase-1,HO1)蛋白表达水平的影响。方法自2020年12月至2021年5月,北部战区总医院烧伤整形科选取40只健康小鼠,其中30只小鼠采用链脲佐菌素腹腔注射法诱导糖尿病小鼠模型成功,小鼠背部做溃疡创面。分组:湿润烧伤膏组(实验组)、康复新液组(阳性对照组)、模型对照组,空白对照组(健康小鼠),每组10只小鼠。实验组和阳性对照组分别给予湿润烧伤膏及康复新液外用换药干预创面,模型对照组和空白对照组不予处理。2周后,采用Western blot法和IHC法检测Nrf2、NQO1、HO1蛋白表达水平,比较4组间的差异。利用凝胶成像分析系统测定目的蛋白灰度值/内参蛋白β-actin灰度值的比值代表该蛋白的相对表达水平,同时数码显微镜下随机视野观察蛋白的平均光密度值作为该蛋白的相对表达水平,并进行对比分析。结果用药2周后实验组、阳性对照组中Nrf2、NQO1、HO1蛋白表达水平高于模型对照组,其差异具有统计学意义(P<0.01);实验组、阳性对照组,模型对照组中Nrf2、NQO1、HO1蛋白表达水平与空白对照组比较,其差异具有统计学意义(P<0.01);蛋白表达电泳图提示,实验组中蛋白表达灰度值/内参蛋白β-actin灰度值的对比最明显,数码显微镜下观察实验组胞浆内Nrf2、NQO1、HO1蛋白表达数量明显增多,4组间差异明显。结论湿润烧伤膏治疗糖尿病小鼠溃疡创面后,Nrf2、NQO1、HO1蛋白表达水平均明显增高。
Objective To investigate the effect of MEBO on protein expression of nuclear factor E2-related factor 2(Nrf2),NADPH quinone oxidoreductase1(NQO1)and heme oxygenase-1(HO1)in muscle tissue of diabetic mice with skin ulcer.Methods From December 2020 to May 2021,40 healthy mice were selected from Department of Burn and Plastic Surgery of General Hospital of Northern Theater Command.Thirty mice were injected intraperitoneally with streptozotocin to induce mouse models of diabetes.The ulcerations were made on the back of mice.All mice were divided into MEBO group(experimental group),Kangfuxin solution group(positive control group),model control group and blank control group(healthy mice),10 mice in each group.The experimental group and positive control group were treated with MEBO and Kangfuxin solution for external dressing intervention.The model control group and blank control group were not treated.After 2 weeks,the protein expression of Nrf2,NQO1 and HO1 were detected by Western blot and IHC.The differences in 4 groups were compared.The ratio of gray value of target protein toβ-actin was detected.The average optical density of protein was observed and analyzed in random field under digital microscope.Results After 2 weeks of treatment,the protein expression levels of Nrf2,NQO1 and HO1 in the experimental group and positive control group were significantly higher than those in the model control group(P<0.01).The protein expression levels of Nrf2,NQO1 and HO1 in the experimental group,positive control group and model control group were significantly higher than those in the blank control group(P<0.01).The ratio of gray value of target protein toβ-actin was most obvious in the experimental group.The protein expression levels of Nrf2,NQO1 and HO1 in the cytoplasm were significantly increased in the experimental group.There were significant differences in 4 groups.Conclusion After the ulcerations treated with MEBO,the protein expression levels of Nrf2,NQO1 and HO1 are significantly increased in diabetic mice.
