摘要
采用荧光定量PCR技术,建立祛痘化妆品中3种致病菌同步快速检验方法,提高检测效率。采用人工染菌方式,离心柱法提取混合模板DNA,设计特异性引物和探针,在同一扩增条件下,完成3种菌同步检测。建立3种菌同时检测荧光定量PCR方法,检测可在24 h内完成,检出限分别为金黄色葡萄球菌1.1×10 CFU/mL,铜绿假单胞菌为1.3×10 CFU/mL,肺炎克雷伯菌为1.3×10 CFU/mL。研究建立的方法灵敏度高、特异性强,可以提高检测效率,建议在祛痘化妆品微生物检测现有标准的基础上作为有效的补充。
A fast detection method based on real-time fluorescence quantitative PCR for three pathogenic bacteria in anti-acne cosmetics was established.The mixed template DNA was extracted with the method of artificial contamination and the spin column method,and the specific primers and probes were designed.Under the same amplification conditions,the simultaneous detection of the three bacteria was completed.Therefore,a fluorescence quantitative PCR method for simultaneous detection of three bacteria was established.The detection can be completed within 24 h.The detection limits are 1.1×10 CFU/mL for staphylococcus aureus,1.3×10 CFU/mL for pseudomonas aeruginosa and 1.3×10 CFU/mL for Klebsiella pneumoniae.The method established in this study has high sensitivity and specificity,which can improve the detection efficiency.
