HPLC法同时测定三叶青不同部位中4种成分含量 被引量:5

Simultaneous determination of four components in different parts of Tetrastigma hemsleyanum Diels et Gilg by HPLC

摘要 目的建立三叶青药材不同部位中4种活性成分的高效液相色谱(HPLC)含量测定方法。方法采用HPLC法测定绿原酸、异槲皮苷、山奈酚-3-O-芸香糖苷、槲皮素4种成分含量。流动相采用乙腈-0.2%磷酸溶液;检测波长:360 nm;色谱柱:Agilent Extend C_(18)柱;柱温:30℃;流速:1.0 ml/min。结果绿原酸、异槲皮苷、山奈酚-3-O-芸香糖苷、槲皮素在1.452~145.2μg/ml,1.245~124.5μg/ml,0.684~68.4μg/ml,1.868~186.8μg/ml内与峰面积呈良好的线性关系(R 2分别为0.9997,0.9995,0.9996,0.9993),平均加样回收率分别为98.8%,101.1%,99.4%,99.2%。三叶青不同部位中4个有效成分的含量存在显著差异,其中三叶青根中山奈酚-3-O-芸香糖苷含量最高,三叶青茎中4种成分含量均在根和叶之间,三叶青叶中绿原酸和槲皮素含量最高但未检测到山奈酚-3-O-芸香糖苷。结论本研究建立的高效液相色谱法能够同时测定三叶青不同部位中4种成分含量,且方法操作简便、快速。 Objective To develop a high performance liquid chromatography(HPLC)method for simultaneously determining four components in different parts of Tetrastigma hemsleyanum Diels et Gilg.Methods The contents of chlorogenic acid,isoquercetin,kaempferol-3-O-rutinoside and quercetin were determined by HPLC.The mobile phase was acetonitrile-0.2%phosphoric acid,the detection wavelength was 360 nm,the chromatographic column was Agilent extend C_(18),the column temperature was 30℃,and the flow rate was 1.0 ml/min.Results Chlorogenic acid,isoquercetin,kaempferol-3-O-rutinoside and quercetin showed a good linear relationship with peak area in the range of 1.452-145.2μg/ml,1.245-124.5μg/ml,0.684-68.4μg/ml,1.868-186.8μg/ml,respectively,and the R 2 were 0.9997,0.9995,0.9996,0.9993,respectively.The recoveries were 98.76%,101.14%,99.39%,and 99.15%,respectively.There were significant differences in the contents of four active components among different parts of Tetrastigma hemsleyanum Diels et Gilg.The content of kaempferol-3-O-rutoside in the root was the highest,the contents of four components in the stem were between the root and the leaf,and the contents of chlorogenic acid and quercetin in the leaf were the highest,but kaempferol-3-O-rutoside was not detected.Conclusion The HPLC method established in this study could simultaneously determine four components in different parts of Tetrastigma hemsleyanum Diels et Gilg,and it is simple and rapid.
