摘要
山东省某规模化养殖场多头猪出现体温升高、咳嗽、呼吸困难、偶尔伴有腹泻症状。为了确诊病因,本试验从病死猪肺脏中分离培养细菌,并对其进行形态学观察、生化鉴定、16S rRNA测序比对、细菌种特异性KMT 1基因鉴定、荚膜血清型鉴定、动物回归试验和药敏试验。结果显示,从患病猪肺脏中分离出的菌株在血平皿上生长出表面光滑且边缘整齐的灰白色菌落,为革兰阴性短杆菌;生化试验显示,该分离菌能发酵蔗糖、葡萄糖等,山梨醇、吲哚、氧化酶、肌醇、甘露醇反应呈阳性;16S rRNA基因核苷酸序列系统进化树分析显示,该分离菌与多杀性巴氏杆菌同源性>99.9%;细菌种特异性KMT 1基因和荚膜血清型鉴定结果显示,该分离菌与多杀性巴氏杆菌荚膜血清D型相符;动物回归试验结果显示,该分离菌有较强的致病性;药敏试验结果显示,该分离菌对氟苯尼考、头孢唑肟、恩诺沙星、链霉素、红霉素和青霉素等9种抗菌药耐药。本试验成功分离到1株猪源荚膜血清D型多杀性巴氏杆菌菌株,可为进一步探索猪多杀性巴氏杆菌病的致病机制和防控提供有效途径。
Pigs in a large-scale farm of Shandong province developed fever,cough,dyspnea and occasionally diarrhea.In order to determine the cause of disease,bacteria were isolated and cultured from the lung of dead pigs,and further examined by morphological observation,biochemical identification,16S rRNA sequence comparison,species-specific KMT 1 gene identification,capsule serotype identification,animal regression test and drug sensitivity test.The results showed that the strain isolated from the lungs of sick pigs produced gray-white colonies with smooth surface and neat edge on blood plate,and was Gram-negative bacillus.Biochemical tests indicated that the isolated strain fermented sucrose and glucose,and was positive for sorbitol,indole,oxidase,inositol,mannitol reactions.Phylogenetic tree of 16S rRNA gene of the isolated strain revealed>99.9%homology with Pasteurella multocida.KMT 1 gene and serotype analyses indicated its P.multocida serotype D identity.Animal regression test demonstrated its strong pathogenicity.The results of drug sensitivity test indicated its resistance to 9 drugs,including florfenicol,ceftizoxime,enrofloxacin,streptomycin,erythromycin and penicillin.Isolation of a P.multocida serotype D strain in this study allows further elucidation of pathogenic mechanism of and development of novel treatment and prevention measures against P.multocida infection in pigs.
