摘要
目的运用网络药理学方法探讨复方丹参滴丸治疗糖尿病心肌病(DCM)的潜在作用机制。方法通过检索TCMSP数据库平台获取复方丹参滴丸活性成分,查询对应靶基因;运用OMIM和GeneCards数据库查找DCM的疾病靶点,构建药物有效成分与疾病靶点的交集Venn图。利用Cytoscape 3.7.2软件构建核心靶点网络图;在DAVID数据库进行GO功能和KEGG通路富集分析。结果筛选获得药物有效成分73个,药-病交集靶点102个,其中IL-6、VEGFA、TNF、TP53等可能是治疗机制核心靶点。富集分析获得GO条目147条,主要包括NO生物合成过程的正调节、质膜、酶结合等生物过程。KEGG通路95条,主要涉及TNF信号途径、癌症信号通路、HIF-1信号通路、PI3K-Akt信号通路等。结论从分子层面分析得到复方丹参滴丸治疗DCM具有多成分、多靶点、多通路的特点,为临床应用提供了一定的理论依据。
Objective To explore the potential mechanism of compound Danshen dropping pills in the treatment of diabetic cardiomyopathy(DCM)by network pharmacology.Methods The active components of compound Danshen dripping pills were obtained by searching TCPSP database platform,and the corresponding target genes were searched.The core target network was constructed by Cytoscape3.7.2 software,and the GO function and KEGG pathway enrichment were analyzed in David Database.Results 73 active components and 102 drug-disease targets were obtained,among which IL6,VEGFA,TNF and TP53 might be the core targets.A total of 147 items of GO were obtained by enrichment analysis,including positive regulation of NO biosynthesis,plasma membrane,enzyme binding and so on.There are 95 KEGG pathways,including TNF pathway,cancer pathway,HIF-1 pathway,PI3K-Akt pathway and so on.Conclusion Compound Danshen dropping pill has the characteristics of multi-component,multi-target and multi-pathway in treating DCM,which provides a theoretical basis for clinical application.
