膀胱感染性结石大鼠的模型构建 被引量:2

Construction of a rat model of bladder infection stones

摘要 目的设计一种新型膀胱感染性结石大鼠模型,模拟出人膀胱刺激症状,作为后续动物实验的研究基础。方法将雄性SD大鼠34只分为模型组(n=14)、无菌水对照组(n=10)和空白对照组(n=10),3组均在麻醉状态下通过金属导丝、推杆将加热塑形的螺旋状异物(聚乙烯材料)置入大鼠膀胱内,模型组给予膀胱灌注脆弱拟杆菌液1×10^(6)个/mL约1.5 mL,无菌对照组给予灌注无菌注射用水约1.5 mL,空白对照组仅按照模型组相同方法放置膀胱异物,不进行灌注。前两组隔天灌注1次,灌注3周,最后一组正常饲养2周。2周后用尿斑实验评估模型组大鼠的排尿行为。实验结束后全部大鼠以颈部脱臼的方法处死,统计各组大鼠膀胱结石率,红外光谱分析结石成分,电镜观察结石表面情况。结果模型组大鼠在灌注1周后死亡2只,其他组大鼠无死亡。实验结束后,模型组的12只存活大鼠膀胱异物均可见结石,与无菌水对照组(3只结石)和空白对照组(2只结石)相比,差异有统计学意义(P<0.05)。各组的结壳成分包括磷酸铵镁和碳酸磷灰石。模型组结石扫描电镜下可见结晶体分层排列,各层次间可见大量细菌生物膜和拟杆菌。模型组的大鼠排尿行为出现异常,表现为尿斑数量增多,尿斑更加靠近滤纸中心区域。结论经大鼠尿道置入膀胱异物并灌注脆弱拟杆菌液可显著提高膀胱结石率,膀胱结石大鼠表现出来的排尿行为存在异常,这与人膀胱刺激症状非常相似。 Objective To design a rat model of bladder infection stones to simulate bladder irritation symptoms for further animal experiments.Methods A total of 34 male SD rats were divided into the model group(n=14),sterile water control group(n=10),and blank control group(n=10).In all 3 groups,heated helical foreign body(polyethylene material)was placed into the rat bladder through a metal guide wire and a push rod under anesthesia.The model group was given intravesical instillation of 1.5 mL Bacteroides fragilis solution 1×10^(6)/mL,the sterile water control group was given 1.5 mL sterile water injection,and the treatment was conducted every other day for 3 weeks.The blank control group was fed normally for 2 weeks.After 2 weeks,the urination behavior of rats in the model group was evaluated with urine spot test.All rats were killed by cervical dislocation.The rate of bladder stones was counted,the stone composition was analyzed with infrared spectroscopy,and the surface of the stones was observed with an electron microscope.Results Two rats in the model group died after 1 week of perfusion,whereas no rats died in the other groups.After the experiment,stones could be observed in 12 rats in the model group,in 3 rats in the sterile water control group,and in 2 rats in the blank control group(P<0.05).The stone components showed magnesium ammonium phosphate and carbonate apatite.Under the scanning electron microscope,stones from the model group had crystals arranged in layers,and a large number of bacterial biofilms and Bacteroides were observed between layers.Rats in the model group exhibited abnormal urination behavior,manifested as an increase in the number of urine spots,which were close to the central area of the filter paper.Conclusion Inserting a foreign body in the bladder through the urethra of the rat and infusion of the Bacteroides fragilis solution can significantly increase the rate of bladder stones.Rats with bladder stones exhibit abnormal urination behavior,similar to the symptoms of human bladder irritation.
