摘要
目的:研究环螺旋蛋白1(NHLH1)在前列腺癌组织中的表达并探讨其与肿瘤治疗和预后的关系。方法:利用GEO数据库、TCGA数据库、Oncomine数据库基因芯片数据筛选出前列腺良恶性组织mRNA差异表达基因NHLH1,统计分析其表达水平与患者生存期的关系;以免疫组化验证前列腺癌组织芯片中NHLH1蛋白表达情况,分析其与临床病理特征的关系;抑制肿瘤细胞株中NHLH1的表达,观察细胞增殖情况。结果:在GSE46602、GSE6956和TCGA数据分析显示,相对于良性组织,NHLH1 mRNA在前列腺癌组织中高表达并与患者生存期有关(P<0.05);免疫组化结果发现,NHLH1蛋白在前列腺癌组织中呈高表达(P<0.05),NHLH1蛋白与Gleason评分和Grade分组显著相关(P<0.05)。结论:NHLH1水平升高是前列腺癌细胞恶性程度升高的标志,预示前列腺癌不良预后,抑制此蛋白表达可减慢肿瘤细胞增殖速度。
Objective:To investigate the expression of nescient helix loop helix 1(NHLH1)in prostate cancer tissue and its relationship with the treatment outcome of prostate cancer.Methods:Gene chip data of GEO database,TCGA database and Oncomine database were used to screen the differential expression of NHLH1 mRNA in benign and malignant prostate tissues.The relationship between its expression level and patient survival was statistically analyzed.Immunohistochemistry was used to verify the protein expression of NHLH1 in prostate cancer tissue microarray.The relationship between expression of NHLH1 protein and clinicopathological features was analyzed.The tumor cell proliferation was studied after suppression of NHLH1 expression.Results:Analyses of GSE46602,GSE6956 and TCGA data sets showed that compared with benign tissues,NHLH1 mRNA was highly expressed in prostate cancer tissues and was associated with patient survival(P<0.05).As shown by immuno⁃histochemistry,NHLH1 protein was highly expressed in prostate cancer tissues(P<0.05).NHLH1 protein was significantly correlated with Gleason score and grading(P<0.05).Conclusion:Elevated NHLH1 is a marker for higher malignancy of prostate cancer cells,and predicts poor prognosis of the cancer.Inhibiting NHLH1 expression can retard the proliferation of tumor cells.
