骨蚀方治疗股骨头坏死的药理学机制 被引量:2

Pharmacological mechanism of Gushi Fang in the treatment of osteonecrosis of femoral head

摘要 目的探讨骨蚀方治疗股骨头坏死(osteonecrosis of the femoral head,ONFH)的潜在药理学机制。方法首先使用TCMSP数据库检索出骨蚀方的活性成分,并根据生物利用度和类药性对检索结果进行筛选,再利用Genecard和OMIM数据库检索出ONFH的疾病靶点,接着使用R语言制作韦恩图得出骨蚀方治疗ONFH的潜在靶标,并用Cytoscape软件构建化合物-靶点-疾病网络,然后运用STRING数据库构建蛋白互作网络图,最后利用R语言进行GO生物功能与KEGG信号通路富集分析。结果骨蚀方中有96个有效活性成分,与ONFH的交集基因121个,信号通路有58个,主要有IL-17、TNF以及P13K-Akt等信号通路参与调控。通过网络图筛选出骨蚀方的关键作用成分为谷甾醇、豆甾醇、槲皮素和β-谷甾醇,关键靶点为IL-6、AKT1、VEGFA、MAPK3和JUN。结论骨蚀方可通过多个基因和信号通路发挥抗炎、调节成骨/破骨细胞比值等作用,进而达到治疗ONFH的效果,体现了骨蚀方多成分、多靶点等特点,也为骨蚀方的临床运用提供了理论依据。 Objective To explore the potential pharmacological mechanism of Gushi Fang in the treatment of osteonecrosis of the femoral head(ONFH).Methods Firstly,TCMSP database was used to retrieve the active ingredients of Gushi Fang,and the search results were screened according to the bioavailability and drug-like properties.Then,the Genecard and OMIM databases were used to retrieve the disease targets of ONFH.Venn map was drawn for potential targets of Gushi Fang in the treatment of ONFH via R language.The compound-target-disease network was drawn by Cytoscape software.The STRING database was used to make a protein interaction network diagram.Finally,GO biological function and KEGG signal pathway enrichment analysis were performed by R language.Results There were 96 active ingredients,121 cross genes with ONFH,and 58 signal pathways in Gushi Fang.IL-17,TNF,and P13KAkt signal pathways were mainly involved in regulation.According to the network diagram,the key components of Gushi Fang were sitosterol,stigmasterol,quercetin andβ-sitosterol,and the key targets were IL-6,AKT1,VEGFA,MAPK3 and JUN.Conclusion Gushi Fang played an anti-inflammatory role and regulated the osteoclast/osteoclast ratio through multiple genes and signaling pathways,thus achieving the therapeutic effect of ONFH,which reflected the characteristics of multi-components and multi-targets of Gushi Fang,and also provided a theoretical basis for clinical application of Gushi Fang.
