摘要
目的建立RP-HPLC法测定胃得安胶囊中23-乙酰泽泻醇B、辛弗林和厚朴酚3种成分的含量研究的分析方法。方法色谱条件:色谱柱为Waters Spherisorb ODS C_(18)柱(250 mm×4.6 mm,5μm);流动相为乙腈(A)-0.5%磷酸溶液(B)、梯度洗脱;检测器为二极管阵列检测器(DAD),采用仪器波长切换功能,分别设定23-乙酰泽泻醇B的检测波长为208 nm,辛弗林的为275 nm,厚朴酚的为294 nm;柱温为30℃;进样量为10μL;流速为1.0 mL/min;二极管阵列检测器定性、定量检测。结果23-乙酰泽泻醇B、辛弗林和厚朴酚分别在0.196~19.66μg/mL、0.299~29.94μg/mL和0.396~39.60μg/mL范围内线性关系良好,平均加标回收率分别为99.4%、99.6%和100.3%。结论所建立的方法具有检测准确度高、操作简便等优点,可用于胃得安胶囊供试品的质量控制。
Objective A RP-HPLC method was established for the determination of alisol B 23-acetate,synephrine and magnolol in Weidean capsules.Methods Waters Spherisorb ODS C_(18)(250 mm×4.6 mm,5μm)chromatographic column was used for separation.The mobile phase consisted of acetonitrile(A)-0.5%phosphoric acid solution(B)with gradient elution at the flow rate of 1.0 mL/min.The diode array detector(DAD)with a wavelength switching function is adopted for qualitative and quantitative analysis,and the detection wavelengths of three components were alisol B 23-acetate(208 nm),synephrine(275 nm),and magnolol(294 nm),respectively.The column temperature was 30℃and the injection volume was 10μL.Results The linearities of alisol B 23-acetate,synephrine and magnolol were in the range of 0.196-19.66μg/mL,0.299-29.94μg/mL and 0.396-39.60μg/mL,respectively.The average recoveries were 99.4%,99.6%and 100.3%,respectively.Conclusion The experimental results showed that the developed method had the advantages of high accuracy and simple operation,and could be used for the quality control of Weidean capsules.
