摘要
目的探讨HEY2和SFRP4在子宫内膜癌(UCEC)中的表达及临床意义。方法通过GEO数据库获取子宫内膜癌表达芯片GSE106191,采用R语言“limma”包进行差异分析,利用GEPIA数据库分析HEY2和SFRP4基因在UCEC中的表达水平,对差异基因进行相关性分析及功能富集分析,利用UALCAN数据库分析子宫内膜癌中的不同临床分期中的表达情况。通过STRING数据库(https://cn.string-db.org/)对低表达基因进行互作分析,并用Cytoscape分析基因间的相关性,选取本院收治的UCEC组织40例作为子宫内膜癌组,同期行子宫切除术的正常子宫内膜组织标本15例为对照组。免疫组化法测定HEY2和SFRP4蛋白的表达,比较两组的表达差异,并采用GEPIA数据库对HEY2和SFRP4在UCEC患者中进行预后分析。结果GEO数据库筛选差异基因,其中HEY2和SFRP4在UCEC中低表达,免疫组化结果与其一致,部分差异基因之间存在显著正相关,且差异基因主要富集在muscle organ development以及乳腺癌等肿瘤相关通路中。HEY2和SFRP4在UCEC不同临床分期与正常组相比具有显著差异,且SFRP4在UCEC患者中低表达组总生存率低。结论UCEC组织中HEY2与SFRP4表达下调,且与预后有关,SFRP4可能作为UCEC有价值的预后标志物。
Objective To explore the expressions and clinical significance of HEY2 and SFRP4 in uterine corpus endometrial carcinoma(UCEC).Methods The endometrial cancer expression microarray GSE106191 was obtained from GEO database,and the limma package of R language was used for differential analysis.The expression levels of HEY2 and SFRP4 genes in UCEC were analyzed by GEPIA database,and the cor⁃relation analysis and functional enrichment analysis of the differential genes were performed.UALCAN data⁃base was used to analyze the expression of endometrial cancer in different clinical stages.The STRING data⁃base(https://cn.string-db.org/)was used to analyze the interaction of low-expressed genes,and Cyto⁃scape was used to analyze the correlation between genes.40 UCEC cases admitted to our hospital were select⁃ed weve taken as endometrial cancer group.15 normal endometrial specimens underwent hysterectomy at the same time as the control group.The expression of HEY2 and SFRP4 protein was measured by immunohisto⁃chemistry,and the difference of expression between the two groups was compared.The prognosis of HEY2 and SFRP4 in UCEC patients was analyzed using GEPIA database.Results The GEO database screened the differential genes,among which HEY2 and SFRP4 were low expressed in UCEC,and the results of immuno⁃histochemistry were consistent with them.There was a significant positive correlation between some differential genes,and the differential genes were mainly enriched in muscle organ development,breast cancer and other tumor related pathways.HEY2 and SFRP4 were significantly different in different clinical stages of UCEC compared with the normal group,and the low expression of SFRP4 in UCEC patients had a low over⁃all survival rate.Conclusion HEY2 and SFRP4 were decreasingly expressed in UCEC tissues,and were as⁃sociated with the prognosis of UCEC,indicating SFRP4 may be served as a potential prognostic biomarker for UCEC.
