银环蛇(Bungarus multicinctus)毒柱层析分离及其组份的活性测定和α-银环蛇毒素的纯化

摘要 本文报导了用CM-Sephadex C-50分离广东产银环蛇毒,得到十三个蛋白组份。测定了九种酶在这些组份中的分布、五个主要毒性组份的神经肌肉阻断作用和小鼠的LD_(50)。小鸡颈二腹肌神经肌肉阻断作用试验表明组分3及组分6显示突触后膜阻断作用,组份9、组份10及组份11为突触前膜阻断作用。对主要的突触后膜毒素组份3进一步用凝胶过滤和离子交换层析进行纯化,得到聚丙烯酰胺凝胶电泳纯一的α-银环蛇毒素。 It is reported in this paper that the Bungarus multicinctus venom from Guangdong Province has been fractionated on CM-Sephadex C-50 into thirteen fractions. Distribution of nine enzymes are determined in the thirteen fractions and neuromuseular blocking action and LD50 of mice (ip) are assayed in five main toxic fractions. Experiment on the neuromuseular blocking action of chick bivenler cervicis muscle showed that fraction 3 and 6 exhibited the property of postsynaplic ncurotoxin (a-type), fraction 9, 10 and 11 contained presynaptic neurotoxin (βtype), as judged from the acetylcholine response of the chick muscle after neuromuseular blockade. Further purification of the fraction 3 by gel filtration on Sephadex G-50 and rechromatography on CM-Scphaclex C-50, purified a-bungarotoxin is obtained. It showed a single band on polyacrylamide gel eleelrophoresis at pH 4.5.
