CST基因家族在乳腺癌中的表达及预后诊断意义

摘要 目的基于在线数据库分析CST基因家族在乳腺癌中的表达及其预后意义,为临床诊疗提供重要指标。方法利用Oncomine数据库获知CSTs在乳腺癌中的转录水平;通过GEPIA、THPA数据库获知CSTs基因家族在乳腺癌中的表达情况;利用Kaplan-Meier Plotter数据库进行生存分析;利用cBioPortal数据库进行乳腺癌CST基因突变情况分析。结果CST1、2、6在乳腺癌中的表达水平高于正常组织(P<0.05)。免疫组化结果显示,与正常乳腺组织比,乳腺癌组织中的CST3蛋白表达呈弱阳性,CST9蛋白表达呈阳性。CST2、3、4、7的高表达在乳腺癌患者中与较好的总体生存率(OS)相关(P<0.05);CSTL1基因高表达与较差的OS相关(P<0.05)。结论CST基因家族大部分成员在乳腺癌中高表达,且CST1、2、3、6、9异常表达,提示其与乳腺癌高度相关;CST2、3、4、7可作为乳腺癌患者预后良好的生物标志物,CSTL1可作为乳腺癌患者预后不良的生物标志物。 Objective Based on online databases,the CST gene family is analyzed for expression in breast cancer and its prognostic significance,providing important indicators for clinical management.Methods The transcript levels of CSTs in breast cancer were obtained from the Oncomine database;the expression of CST gene family in breast cancer was obtained from the GEPIA and THPA databases;the Kaplan-Meier Plotter database was used for survival analysis;and the cBioPortal database was used for the analysis of CST gene mutations in breast cancer.Results The expression levels of CST1,2,and 6 were higher in breast cancer than in normal tissues(P<0.05).Immunohistochemical results showed that CST3 protein expression was weakly positive and CST9 protein expression was positive in breast cancer tissues compared with normal breast tissues.High expression of CST2,3,4,and 7 was associated with better overall survival(OS)in breast cancer patients(P<0.05);high CSTL1 gene expression was associated with poorer OS(P<0.05).Conclusion Most members of the CST gene family are highly expressed in breast cancer,and CST1,2,3,6,and 9 are aberrantly expressed,suggesting that they are highly associated with breast cancer;CST2,3,4,and 7 can be used as biomarkers for good prognosis in breast cancer patients,and CSTL1 can be used as a biomarker for poor prognosis in breast cancer patients.
