摘要
目的:通过网络药理学探讨当归四逆汤中“当归-桂枝”药对对治疗痛经的可能作用机制。方法:通过中药系统药理学平台(TCMSP)检索当归四逆汤中当归与桂枝的化学成分和靶点,再通过Drug Bank数据库查询痛经相关靶点,进而构建化合物-疾病靶点网络和蛋白-蛋白相互作用(PPI)网络,进行基因功能富集分析和通路富集分析,并构建靶点-通路网络。结果:在中药当归中共筛选出潜在活性成分2个,桂枝活性成分7个,二者活性成分的作用靶点分别为69个和74个,其中作用于痛经的相关靶点有6个,涉及PGR、 PTGS1、 PTGS2等。化合物-疾病靶点网络共包含了7个化合物和对应靶点6个,关键化合物涉及β-谷固醇等。PPI网络包含6个节点,关键的作用靶点共涉及PTGS2、 ESR1、PGR等。基因本体(GO)富集分析得到条目4个,其中包括生物过程的相关条目有2个,与分子功能相关的条目共有2个。KEGG通路5条,涉及脂肪细胞中脂肪分解的调节、花生四烯酸的代谢和利什曼病等。结论:基于网络药理学的研究方法初步验证了当归-桂枝药对其治疗痛经的作用机制,并为进一步深入研究其作用机理奠定了良好的基础。
Objective Using the method of network pharmacology to study the mechanism of Angelicae Sinensis Radix-Cinnamomi Ramulus in Danggui Sini Decoction on dysmenorrhea.Methods The chemical constituents and targets of Angelica sinensis and Radix-Cinnamomi Ramulus in Danggui Sini Decoction were retrieved through the Traditional Chinese Medicine System Pharmacology Platform(TCMSP),and then the related targets of dysmenorrhea were queried through the DrugBank database.The Compound-Disease target network and the Protein-Protein Interaction network were constructed respectively.Finally,gene function enrichment analysis and pathway enrichment analysis were performed,and a target-pathway network was constructed.Results 2 candidate active molecules and 69 corresponding targets in Angelicae Sinensis Radix and 7 candidate active molecules and 74 corresponding targets in Cinnamomi Ramulus were screened out.There were 6 targets related to dysmenorrhea,involving PGR,PTGS1,PTGS2,etc.The compound-disease related target network contained 7 chemical components and 6 targets,in which the hub components involved Beta-sitosterol,etc.The PPI network contained 6 nodes,in which the hub targets involved PTGS2,ESR1,PGR,etc.In GO functional enrichment analysis,there were 4 GO terms(2 Biological Process and 2 Molecular Function).In KEGG pathway enrichment analysis,there were 5 KEGG pathways,involving regulation of lipolysis in adipocytes,Arachidonic acid metabolism,leishmaniasis,etc.Conclusio ns Using the research method of network pharmacology,the action mechanism of Angelicae Sinensis Radix and Cinnamomi Ramulus in the treatment of dysmenorrhea was preliminarily verified,and a good foundation was laid for further in-depth study of its action mechanism.
