杉木组培苗生根配方的筛选及对内源激素的影响 被引量:4

摘要 [目的]筛选适合杉木‘洋020’组培苗生根的培养基配方,并从激素角度了解组培苗生根调控机理,为杉木组培苗工厂化生产提供参考。[方法]以杉木优良无性系组培苗‘洋020’为试验材料,比较不同浓度MS培养基(1×、0.50×、0.25×)、不同生长素(IBA和NAA)浓度及其配比对组培苗生根的影响,筛选出适合‘洋020’的生根配方。测定初筛试验中生根率最低处理(T_(1))和优化试验中生根率最高处理(M_(5))的茎尖内源激素含量。[结果](1)优化后筛选出适宜‘洋020’组培苗生根的配方(M_(5))为:0.25×MS+0.30 mg·L-1 IBA+0.075 mg·L^(-1)NAA,生根率达85.93%。(2)与T 1处理相比,M 5处理在生根高峰期(21 d后)维持较高水平的IAA、GA、ABA和ZR含量,以及较低水平的CTK含量。(3)与T_(1)相比,M_(5)处理在生根中前期(21 d前)维持较低水平的IAA/GA、IAA/ABA、IAA/CTK和IAA/ZR比值,而在生根后期(21 d后)维持较高水平的IAA/ABA、IAA/GA和IAA/CTK比值。[结论]0.25×MS+0.30 mg·L^(-1)IBA+0.075 mg·L^(-1)NAA是适宜杉木优良无性系‘洋020’生根的配方。添加适宜浓度的IBA和NAA可以提高组培苗生根率,可能与在一定程度上改变了组培苗内源激素含量及其比值有关。 [Purpose]Screening suitable rooting medium formula for Cunninghamia lanceolata tissue culture seedlings of‘Yang 020’,and revealing the regulation mechanism for rooting from the perspective of hormones could provide reference for the industrialized production of C.lanceolata tissue culture seedlings.[Method]The effects of different MS culture media types(1×、0.50×、0.25×)and auxin(IBA and NAA)with various concentrations and ratios on the rooting of tissue culture seedlings were compared in order to select the most suitable rooting formula for‘Yang 020’.The differences of endogenous hormone contents in shoot tips were analyzed between the lowest rooting rate in the initial screening test(MS medium without auxin,T_(1))and the highest rooting rate in the optimized experiment(M_(5))during rooting process.[Result](1)The rooting formulas suitable for‘Yang 020’tissue culture seedlings were selected after optimized experiment:0.25×MS+0.30 mg·L^(-1)IBA+0.075 mg·L^(-1)NAA,with the rooting rate of 85.93%.(2)Compared with T_(1) treatment,higher levels of IAA,GA,ABA and ZR contents and lower levels of CTK contents were observed in M_(5) treatment at the rooting peak(after 21 d).(3)Compared with T_(1) treatment,lower levels of IAA/GA,IAA/ABA,IAA/CTK and IAA/ZR ratios were observed in M_(5) treatment in the middle and early stages of rooting(before 21 d),while higher IAA/ABA,IAA/GA and IAA/CTK ratios were present in the late stage of rooting(after 21 d).[Conclusion]0.25×MS+0.30 mg·L^(-1) IBA+0.075 mg·L^(-1) NAA is the formula suitable for rooting of superior clone‘Yang 020’.Adding appropriate concentration of IBA and NAA could improve the rooting rate of tissue culture seedlings,which maybe related to the change of endogenous hormone content and its ratio to a certain extent.
