摘要
[目的]探讨端锚聚合酶(TNKS)在消岩汤抗肺腺癌细胞作用中的表达及意义。[方法]采用免疫组织化学检测37例肺腺癌患者癌及癌旁组织中TNKS的表达情况。实时荧光定量聚合酶联反应(qRT-PCR)检测梯度浓度消岩汤处理肺腺癌A549细胞后,TNKS mRNA水平的表达;CCK-8法、γ-H2AX细胞免疫荧光、DNA修复试验分别检测各组A549细胞的增殖、DNA损伤及DNA修复能力,划痕实验及Transwell小室分别检测各组A549细胞的迁移及侵袭能力。[结果]1)TNKS蛋白在肺腺癌组织中的阳性表达率为63.00%,在癌旁组织中阳性表达率为18.92%,差异有统计学意义(P<0.05)。2)随消岩汤浓度升高,A549细胞中TNKS mRNA水平的表达降低。3)当TNKS被下调时,A549细胞增殖活性降低,γ-H2AX表达增强,细胞DNA修复能力、迁移和侵袭能力受抑制,同样给予消岩汤处理细胞后,结果与TNKS下调组一致。[结论]TNKS与肺腺癌细胞的增殖、DNA损伤修复、迁移和侵袭密切相关,消岩汤抗肿瘤作用可能与下调TNKS的表达有关。
[Objective]To investigate the expression and significance of tankyrases(TNKS)in Xiaoyan Decoction against lung adenocarcinomacells.[Methods]The expression of TNKS in lung adenocarcinoma tissues and adjacent tissues was detected by immunohistochemistry in 37 cases.qRT-PCR was used to detectthe mRNA expression of TNKS in A549 cells treated with gradient concentration of Xiaoyan Decoction.CCK-8 assay,γ-H2AX immunofluorescenceand DNA repair assay were used to detect the cell proliferation,DNA damage and DNA repair respectively in each group of A549 cells.The scratch test and transwell chambers were used to detect the migration and invasion capacity of cells in each group of A549 cells.[Results]1)The positive expression rates of TNKS protein inlung adenocarcinoma tissues and paracancerous tissues were 63.00%and 18.92%,respectively.The differences were statistically significant(P<0.05).2)The mRNA expression of TNKS in A549 cells decreased with the increase of Xiaoyan Decoction concentration.The differences were statistically significant(P<0.05).3)After TNKS was down-regulated,the cell proliferative activity was decreased and the expression ofγ-H2AX was increased.The ability of DNA repair,cell migration and invasion were all inhibited.Similarly,after cells were treated with Xiaoyan Decoction,the results were consistent with those in the TNKS down-regulated group.[Conclusion]TNKS are closely related to cellproliferation,DNA damage and repair,migration and invasion of lung adenocarcinomacells.The antitumor effect of Xiaoyan Decoction may be related to the down-regulation of TNKS expression.
