摘要
为了建立一种类似2型糖尿病患者的动物模型,采用饲喂高糖高脂饲料和注射小剂量(40 mg/kg)链脲佐菌素(STZ)的方法模拟建立2型糖尿病小鼠模型。将80只雄性C57 BL/6J小鼠随机分配为2型糖尿病(T2D)组和正常对照(CHOW)组,分别以高糖高脂饲料和基础饲料喂养后观察两组小鼠体重和血糖的变化。结果发现,T2D组小鼠的体重和血糖均高于CHOW组且趋于稳定,此外T2D组小鼠表现出明显的葡萄糖耐量异常和胰岛素抵抗现象;通过对两组小鼠的肝脏和脂肪组织进行苏木精-伊红染色发现,T2D组小鼠两种组织中的脂肪泡均明显多(大)于CHOW组。因此采用饲喂高糖高脂饲料和连续3 d腹腔注射40 mg/kg剂量的STZ的方法诱导建立2型糖尿病小鼠模型,成模率和存活率达80%以上,说明该模型的建立方法经济实用且方便有效。
In this study,the model of type 2 diabetes was established by feeding high glucose and high lipid diet and injecting a small dose(40 mg/kg)of streptozotocin(STZ).A total of 80 male C57 BL/6J mice were randomly assigned to a type 2 diabetes mellitus(T2D)group and a normal control(CHOW)group.The mice in the two groups were fed with high glucose and high lipid diet or basic diet,respectively.The changes of body weight and blood glucose were observed.The results showed that the body weight and blood glucose of mice in the T2D group were higher than those in the CHOW group and remained stable.In addition,mice in the T2D group showed significant impaired glucose tolerance and insulin resistance.The results of hematoxylin and eosin staining of liver and adipose tissues from the two groups of mice showed that the fat vacuoles in both tissues of the T2D group were significantly larger than those in the CHOW group.Therefore,the type 2 diabetes mice model was induced by feeding with high glucose and high lipid diet and intraperitoneal injection of 40 mg/kg STZ for three consecutive days,and the modeling rate and survival rate reached over 80%,indicating that the model establishment method was economical,practical,convenient and effective.
