摘要
建立了测定化妆品中虾青素含量的高效液相色谱方法。样品经乙腈提取,0.1 mol/L NaOH-甲醇溶液皂化反应后,C18色谱柱(250 mm×4.6 mm,5μm)分离,以甲醇-水(95∶5,V/V)洗脱,采用高效液相色谱法(HPLC)检测。对流动相条件进行优化,实现了全反式、9-顺式和13-顺式虾青素的有效分离,且用质谱对同分异构体结构进行了鉴定;对前处理方法进行考察,验证了皂化反应的必要性并优化了反应条件。虾青素在0.1~10μg/mL范围内呈良好的线性关系,相关系数为0.9992;在5,10,20μg/g 3个加标水平下,在液态水基和膏霜乳液类化妆品中的回收率为82.1%~107.5%,相对标准偏差(RSD)为2.1%~3.7%,检出限(LOD)分别为0.57和0.55μg/g,定量限(LOQ)分别为1.9和1.8μg/g。该方法适用于化妆品中虾青素的检测。
A method of high performance liquid chromatography(HPLC)was established for the determination of astaxanthin in cosmetics. The sample was extracted with acetonitrile solution,and separated on a C18 column(250 mm×4.6 mm,5 μm)with methanol-water(95∶5,V∶V)as mobile phases. After optimizing the mobile phase conditions,all-trans,9-cis and 13-cis-astaxanthin were completely separated,and the structures of isomers were identified by mass spectrometry(GC). The pretreatment method was investigated to verify the necessity of saponification reaction and the conditions of saponification reaction were optimized. Results indicated that astaxanthin had a good linear relationship in the range of 0.1-10 μg/mL,and the correlation coefficient was0.9992. Average recoveries of astaxanthin in toner and cream cosmetics ranged from 82.1% to 107.5% at three spiked levels of 5,10 and 20 μg/g and the relative standard deviations(RSDs)were 2.1%-3.7%.The limits of detection(LODs)were 0.57 and 0.55 μg/g and the limits of quantification(LOQs)were 1.9 and 1.8 μg/g,respectively. The method is suitable for the detection of astaxanthin in cosmetics.
