基于数据挖掘探讨中药治疗卵泡发育不良用药规律及实验验证 被引量:4

Medication Rule of Chinese Medicine for Treatment of Follicular Dysplasia Based on Data Mining and Experimental Validation

摘要 目的:数据挖掘中药治疗卵泡发育不良的用药规律,并探讨聚类分析所得中药新方对排卵障碍大鼠卵巢功能的影响。方法:检索近20年相关中英文文献,提取、整理中药信息,采用SPSS Modeler18.0、SPSS Statistics 24.0软件分析中药治疗卵泡发育不良的用药特点、关联规则。确定聚类分析所得中药新方用于实验研究,将SD雌性大鼠随机分为对照组、模型组(雄激素致排卵障碍大鼠模型)、来曲唑组(模型组+来曲唑)、补肾健脾方组(模型组+补肾健脾方),每组10只。苏木精-伊红(HE)染色观察大鼠卵巢内各级卵泡发育情况、酶联免疫吸附试验检测大鼠血清雌二醇(E_(2))、促黄体素(LH)、促卵泡素(FSH)、孕激素(P)等性激素水平。结果:研究纳入132个方剂,包含146味中药,涉及肾虚血瘀证、肾阴虚证等8个证型,使用频次≥20的有当归、熟地黄、菟丝子等27味中药,药味以甘、辛为主,药性以温、平为主,归经以肾为主,脾、肝次之。关联规则强度最高药对为当归、熟地黄-菟丝子,聚类分析获得三类新药组合,经专家讨论新方“补肾健脾方”用于实验研究。结果显示雄激素所致排卵障碍大鼠模型卵巢内大多卵泡呈囊性扩张,几乎无黄体生成。补肾健脾方组大鼠卵巢内发育卵泡增多,颗粒细胞及黄体数目增加,且血清E_(2)、P及FSH水平均有所升高。结论:通过数据挖掘中药治疗卵泡发育不良以补肾药物为主,聚类分析所获补肾健脾方可改善大鼠卵巢组织形态,促进卵泡发育,并改善血清激素水平,有效提高卵巢储备功能。 Objective:To explore the medication rule of Chinese medicine in treating follicular dysplasia by data mining,and the effect of new Chinese medicine prescriptions obtained by cluster analysis on ovarian function in rats with ovulation disorder.Methods:Relevant Chinese and English literature in the past 20 years were retrieved.The data of Chinese medicine were extracted and sorted out,and the medication characteristics and association rules of Chinese medicine for the treatment of follicular dysplasia were analyzed using SPSS Modeler 18.0 and SPSS Statistics 24.0.New Chinese medicine prescriptions obtained by cluster analysis was determined for experimental research.Female SD rats were randomly divided into control group,model group(rat model of androgen-induced ovulation disorder),Letrozole group(model group+Letrozole),and Bushen Jianpi Prescription group(model group+Bushen Jianpi Prescription),with 10 in each group.Hematoxylin and eosin(HE)staining was used to observe the development of follicles at all levels in the ovaries of rats,and enzyme-linked immunosorbent assay(ELISA)was used to detect the levels of serum estradiol(E_(2)),luteinizing hormone(LH),follicle stimulating hormone(FSH),progesterone(P)and other sex hormones.Results:A total of 132 prescriptions were included in the study,involving 146 Chinese herbal medicines and treating 8 syndromes such as kidney deficiency and blood stasis syndrome and kidney yin deficiency syndrome.Among the Chinese herbal medicines,27 such as Radix Angelicae Sinensis,Radix Rehmanniae Preparata,and Semen Cuscutae were used more than 20 times.They were mainly sweet and pungent in taste and warm and mild in property,acting on kidney,followed by spleen and liver.The herbal pairs Radix Angelicae Sinensis-Radix Rehmanniae Preparata and Radix Rehmanniae Preparata-Semen Cuscutae had the highest reliability of association rules,and three new combinations were obtained by cluster analysis.After expert discussion,the new formula Bushen Jianpi Prescription was used for experimental research.The results showed that most of the follicles in the ovaries of the rats with androgen-induced ovulation disorder had cystic expansion,and there was almost no luteinization.In contrast,the number of developing follicles,granulosa cells and corpus luteum in the ovaries of the rats in the Bushen Jianpi Prescription group increased,and the levels of serum E_(2),P and FSH were up-regulated.Conclusion:Through data mining,Chinese medicines for tonifying kidney are mainly used in the treatment of follicular dysplasia.Bushen Jianpi Prescription obtained by cluster analysis improves the ovarian tissue morphology,promotes follicular development,and regulates serum hormone levels,thereby enhancing ovarian reserve function in rats.
