蓼子朴核型分析

摘要 本试验采用一般染色体制片技术,对蓼子朴水培生长的新根进行预处理、固定、制片、镜检以及拍照等过程,获得清晰的含中期染色体的图片后进行核型分析。结果表明:塔里木盆地周边的蓼子朴染色体数目是2n=2x=16,染色体相对长度由2n=16=4L+2M2+6M1+4S组成,蓼子朴核型公式为K(2n)=2x=16=14m(2SAT)+2sm,最长染色体与最短染色体的比值为2.41,臂比值大小在1.07~2.88之间,12.5%的染色体臂比值大于2,在核型分类上属于2B,且核型不对称系数(AS.K%)为53.9%。由此可知,蓼子朴的核型属于由对称向不对称演化的过渡类型,对蓼子朴核型分析有助于解决沙生植物驯化栽培、降低沙漠化等生态问题,为进一步确定其进化程度及遗传工程等奠定基础。 In this experiment,the general chromosome production technology was used to carry out pretreatment,fixation,production,microscopic examination,photographing and other processes on the new roots of I.salsoloides.After obtaining clear pictures containing metaphase chromosomes,karyotype analysis was carried out.The results show that the relative length of the chromosome is 2n=16=4L+2M2+6M1+4S,and the karyotype formula of I.salsoloides is K(2n)=2x=16=14m(2SAT)+2sm,the ratio of the longest chromosome to the shortest chromosome is 2.41,the ratio of arms is between 1.07 and 2.88,the ratio of arms is greater than 2 in 12.5%of the chromosomes,and it belongs to 2B in karyotype classification and nuclear asymmetry coefficient(AS.K%)is 53.94%.It can be seen that the karyotype of I.salsoloides belongs to the transition type from symmetry to asymmetry.The karyotype analysis of I.salsoloides is helpful to solve the ecological problems such as the domestication and cultivation of desert plants and the reduction of desertification,and lays a foundation for further determining its evolution degree and genetic engineering.
