牙龈卟啉单胞菌对牙周炎患者成骨分化及炎性因子的影响

Effect of Porphyromonas Gingivalis on Osteogenic Differentiation and Inflammatory Factors in Patients with Periodontitis

摘要 目的:探讨感染牙龈卟啉单胞菌对牙周炎患者成骨分化及白介素-6(IL-6)、白介素-8(IL-8)、肿瘤坏死因子-α(TNF-α)水平的影响。方法:选取2020年1月-2021年4月北京华信医院收治的感染牙龈卟啉单胞菌牙周炎患者45例为观察组,同期选择健康人群45例为对照组,进行人牙周膜细胞(HPDLCS)培养。比较两组HPSLCS增殖活性、成骨分化、Runx2、IL-6、IL-8、TNF-α水平。结果:1~7 d,两组HPDLCS细胞增殖情况比较,差异无统计学意义(P>0.05);对照组经茜素红染色结果显示成骨诱导后出现较为明显的矿化结节,成骨分化良好,观察组矿化结节明显减少,成骨分化不良;观察组Runx2的mRNA表达量低于对照组,IL-6、IL-8、TNF-α细胞炎性因子表达量高于对照组,差异有统计学意义(P<0.05)。结论:感染牙龈卟啉单胞菌对HPDLCS增殖无明显影响,但可抑制细胞成骨分化,促进炎性因子分泌、炎性反应的发生。 Objective:To investigate the effects of infection with Porphyromonas gingivalis on osteogenic differentiation and levels of interleukin-6(IL-6),interleukin-8(IL-8),and tumor necrosis factor-α(TNF-α)in patients with periodontitis.Methods:45 patients infected with Porphyromonas gingivalis periodontitis admitted to Beijing Huaxin Hospital from January 2020 to April 2021 were selected as the observation group.During the same period,45 cases of healthy population were selected as the control group,and human periodontal membrane cells(HPSDLCS)culture was performed.The proliferative activity,osteogenic differentiation,Runx2,IL-6,IL-8,and TNF-αlevels of HPSLCS were compared between the two groups.Results:From 1 to 7 d,there was no statistically significant difference in the proliferation of HPDLCS cells between the two groups(P>0.05).The control group showed more obvious mineralized nodules and good osteogenic differentiation after osteogenic induction by alizarin red staining results.In the observation group,mineralized nodules were significantly reduced and osteogenic differentiation was poor.The mRNA expression of Runx2 in the observation group was lower than that in the control group,and the expression of IL-6,IL-8 and TNF-αcellular inflammatory factors was higher than that in the control group,and the difference was statistically significant(P<0.05).Conclusion:Infection with Porphyromonas gingivalis had no significant effect on the proliferation of HPDLCS,but it could inhibit cellular osteogenic differentiation and promote the secretion of inflammatory factors and inflammatory response.
