HA-SA复合微凝胶的制备与评价

摘要 以透明质酸钠(HA)和海藻酸钠(SA)为原料,利用碳二亚胺体系催化HA与己二酸二酰肼(ADH)发生化学交联,以及SA与Ca^(2+)发生物理交联,并结合乳化法制备了一种具有互穿网络结构的HA-SA复合微凝胶.通过形貌特征分析、红外光谱(IR)、X射线衍射(XRD)分析、溶胀测试等一系列表征对HA-SA复合微凝胶进行了物理化学性能考察,并探索了复合微凝胶的最佳释药性能.结果表明:当HA和ADH的摩尔比为1∶1,HA浓度为2%、SA浓度为4%时,复合微凝胶溶胀率可达44;Ca^(2+)浓度为2.5%,交联时间为12 h时,复合微凝胶有最优的释药性能,且对模型药物姜黄素的缓释效果要优于吲哚美辛和二甲双胍.该复合微凝胶具有明显pH敏感性且可控制药物于模拟肠液中持久释放,在药物递送和治疗肠道疾病领域上具有潜在价值. By using the chemical crosslinking between sodium hyaluronate(HA)and adipic dihydrazide(ADH)catalyzed by carbodiimide system,and the physical crosslinking between sodium alginate(SA)and Ca^(2+),the HA-SA composite microgels with interpenetrating network structure was fabricated.Through a series of characterizations such as morphology,infrared spectroscopy(IR),X-ray diffraction(XRD),swelling test,etc.,the physical and chemical properties of HA-SA microgels were investigated,and the optimal behavior of drug release was explored.The results show that the swelling rate of the composite hydrogel was 44 as the molar ratio of HA and ADH is 1∶1,the HA concentration is 2%,and the SA concentration is 4%;once the Ca^(2+)concentration is 2.5%,and the cross-linking time is 12 h,the composite microgels showed the best drug release performance.The release behavior indicated that the sustained release effect of the composite microgels for curcumin was better than that of indomethacin and metformin.The composite microgels exhibits obvious pH sensitivity and sustained drug release behavior in the simulated intestinal fluid,and it has potential value in the field of drug delivery and treatment of intestinal diseases.
